Expanding the functionality of proteins with genetically encoded dibenzo[b,f][1,4,5]thiadiazepine: a photo-transducer for photo-click decoration

Summary: This study presents a method for genetically encoding proteins with novel noncanonical amino acids, enabling site-specific functionalization of proteins in living cells through photo-click reactions. By designing and synthesizing a photo-switchable amino acid and using directed evolution to encode it into recombinant proteins, a genetically encodable reporter for photo-click bioconjugation with spatiotemporal controllability is developed. The photo-isomerization of the amino acid can be used as a tool to convert photon energy into ring-strain energy, oscillating the conformational changes of the parent proteins. The photo-switching of the amino acid on proteins enables efficient and selective photo-click ligation with specific probes. This genetic code expansion method allows for further functional diversity of artificial proteins in living systems.

A General Strategy to Design Highly Fluorogenic Far-Red and Near-Infrared Tetrazine Bioorthogonal Probes

Summary: A novel strategy has been developed to create highly fluorescent Huaxi-Fluor probes that exhibit significantly increased fluorescence upon forming the bioorthogonal reaction product, pyridazine. These probes have large Stokes shifts and high quantum yields, with emission wavelengths fine-tunable from 556 to 728 nm by manipulating the fluorophore skeleton. Highly photo-stable and biocompatible, these probes are suitable for live cell organelle visualization without washing steps and imaging tumors in live small animals to depths of 500 μm by two-photon excitation.
