Hopf bifurcation mechanism analysis in an improved cortex-basal ganglia network with distributed An to Parkinson's disease

Zhizhi Wang et al.

Summary: In this paper, the authors investigate the mechanisms of Hopf bifurcation for Parkinson's oscillations by introducing general distributed delays and self-feedback distributed delays into an improved cortex-basal ganglia network. The results show that general distributed delays inhibit oscillations in the basal ganglia, while self-feedback distributed delays promote oscillations. The effects of strong and weak coupling kernels on Hopf bifurcations mainly depend on the value of self-feedback distributed delays. The authors define four different states in the model to explain the different origins of oscillations. The increase of distributed delays can induce the transition between supercritical and subcritical Hopf bifurcations, leading to changes in oscillation frequency bands.

Driving motor cortex oscillations modulates bradykinesia in Parkinson's disease

Andrea Guerra et al.

Summary: In this study, the authors found that abnormal beta and gamma oscillations in the basal ganglia are related to bradykinesia in patients with Parkinson's disease. Transcranial alternating current stimulation (tACS) can modulate cortical rhythms and was found to affect movement speed and amplitude in this study. Changes in inhibitory interneuronal activity may reflect compensatory mechanisms in the primary motor cortex (M1) to counteract bradykinesia.

A bidirectional Hopf bifurcation analysis of Parkinson's oscillation in a simplified basal ganglia model

Bing Hu et al.

Summary: In this paper, the oscillation mechanism in a computational model of Parkinson's disease was studied using bifurcation analysis and numerical simulation. The findings showed that abnormal coupling weights and delays can induce oscillatory activities. The study identified bidirectional Hopf bifurcations that can explain the oscillation mechanism in the model. Furthermore, it was observed that the effect of delay in inhibitory pathways is greater than that in excitatory pathways.

Hopf bifurcation analysis for Parkinson oscillation with heterogeneous delays: A theoretical derivation and simulation analysis

Zhizhi Wang et al.

Summary: In this paper, the origin mechanism, oscillation amplitude, and frequency characteristics of Parkinson's oscillation in an improved cortex-basal ganglia resonance model (EXN-INNSTN-GPe) were systematically studied. The study found that the delay is a key factor affecting the oscillation frequency, and various frequency bands appear with an increase in delay. The STN-GPe loop can be divided into four types of states based on critical conditions derived from heterogeneous delays, representing different origin mechanisms of oscillation. The amplitude characteristics vary with different types, and the induced oscillation amplitude is generally smaller than the spontaneous oscillation amplitude. The study also revealed the important roles of self-feedback connections in affecting the amplitude of induced oscillation. The theoretical analysis is in good agreement with numerical simulation, providing valuable insights for experimental and theoretical research in Parkinson's disease.

Pallidal Connectivity Profiling of Stimulation-Induced Dyskinesia in Parkinson's Disease

Takashi Tsuboi et al.

Summary: This study aimed to identify anatomical regions related to stimulation-induced dyskinesia (SID) in PD patients after pallidal DBS and analyze the connectivity associated with SID. The results showed that SID VTAs were primarily located in the dorsal GPi/GPe and had higher structural connectivity to certain cortical regions compared to non-SID VTAs. Differences in connectivity between motor-related cortices and subcortical regions may explain the presence or absence of SID.

Primary motor cortex in Parkinson's disease: Functional changes and opportunities for neurostimulation

Conor F. Underwood et al.

Summary: Movement abnormalities in Parkinson's disease are caused by disordered neural activity in interconnected brain structures, particularly affecting the recruitment of pyramidal projection neurons in the primary motor cortex. Alterations in M1 function in PD can be improved with neurostimulation methods that directly or indirectly modulate M1 activity, showing promise for targeted neuromodulation as a therapy for PD patients.

Connectivity and Functionality of the Globus Pallidus Externa Under Normal Conditions and Parkinson's Disease

Jie Dong et al.

Summary: The globus pallidus externa (GPe) serves as a central hub in the basal ganglia system for processing motor and non-motor information, forming complex connections with other nuclei and brain regions. Recent advances in genetic tools have led to a better understanding of the molecular, anatomical, electrophysiological, and functional properties of GPe neurons and non-neuronal cells. Impairments in dopamine transmission in the basal ganglia, which contribute to Parkinson's disease (PD), are associated with altered GPe neuron activity and synaptic connections, as observed in both PD patients and pre-clinical models.
