Accurate de novo design of membrane-traversing macrocycles

Gaurav Bhardwaj et al.

Summary: This study investigates the design principles for macrocycle membrane permeability and oral bioavailability using computational design and experimental characterization. The results show that membrane permeability can be achieved by engaging all amide (NH) groups in internal hydrogen bonding interactions. Additionally, designs with exposed NH groups can be made membrane permeable through the design of an alternative isoenergetic fully hydrogen-bonded state favored in the lipid membrane. The ability to design membrane-permeable and orally bioavailable peptides with high structural accuracy has significant implications for the development of next-generation macrocycle therapeutics.

Artificial channels for confined mass transport at the sub-nanometre scale

Jie Shen et al.

Summary: This review discusses the design, mass transport mechanisms, and potential applications of artificial channels that selectively transport small molecules at the sub-nanometer scale. It highlights the platforms of well-defined nanopores, 1D nanotubes, and 2D nanochannels enabled by synthetic nanomaterials, and explores their structure-property-performance relationships. The practicalities and potential impact of these channels on next-generation sustainable technologies for energy, the environment, and healthcare applications are also discussed.

Nanotube Active Water Pump Driven by Alternating Hydrophobicity

Noriyoshi Arai et al.

Summary: Efficient control of water transport is essential for the future sustainability of life. Various water transport systems utilizing carbon nanotubes have been proposed, but in this study, a novel active water pump driven by hydrophobic interaction was successfully demonstrated through computer simulation. This pump can actively transport water molecules by alternately adjusting the hydrophobicity of its surface, offering potential for applications in low-energy artificial purification systems.

Terahertz Wave Enhances Permeability of the Voltage-Gated Calcium Channel

Yangmei Li et al.

Summary: The dysfunction of voltage-gated calcium channels leading to a deficiency in Ca2+ fluxes is associated with calcium channelopathies such as epilepsy and periodic paralysis. By using resonant terahertz waves, researchers have demonstrated enhanced selectivity and conductance of Ca2+, potentially offering therapeutic interventions in correcting calcium channel deficiencies and inducing apoptosis in tumor cells.

Controlling Water Flow through a Synthetic Nanopore with Permeable Cations

Yi Shen et al.

Summary: Researchers have developed a self-assembling nanopore with a hybrid hydrophilic/hydrophobic lumen that allows for regulated water transport by alkali metal ions. The study demonstrates a non-monotonic dependence of water transport on cation size, with a broad range of water flow observed, from minimal in the presence of sodium ions to levels comparable to a biological water channel in their absence. Molecular dynamics simulations reveal that the sensitivity of water flow is due to the binding of cations within the pore, providing a proof-of-concept for the rational design of controllable synthetic water channels.
