Nitrogenation of Amides via C-C and C-N Bond Cleavage

Ming -Hui Zhu et al.

Summary: Amides are important chemicals in organic chemistry. Converting amides to other functional products is challenging, but a new method using a direct nitrogenation approach has been developed. This method utilizes C-C and C-N bond cleavage and employs the inexpensive inorganic salt NaNO2 as a nitrogen source. Applications of this method include modifying drugs and natural product derivatives.

Facile preparation of dihydro-1,4-benzothiazine derivatives via oxidative ring-expansion of 2-aminobenzothiazoles with olefins

Qing Sun et al.

Summary: A concise and efficient method for the preparation of dihydro-1,4-benzothiazine derivatives via oxidative ring-expansion of 2-aminobenzothiazoles with olefins under metal-free conditions is described. The protocol demonstrates moderate-to-good yields and is applicable to a wide range of readily accessible 2-aminobenzothiazoles and olefins. The [4+2] heteroannulation between the intermediacy of oxidative ring-opening of 2-aminobenzothiazoles and olefins is suggested to rationalize the formation of the product.

Palladium-NHC (NHC = N-heterocyclic Carbene)-Catalyzed Suzuki-Miyaura Cross-Coupling of Alkyl Amides

Chang-An Wang et al.

Summary: We report the Pd-catalyzed Suzuki-Miyaura cross-coupling of aliphatic amides. The selective N-C(O) cleavage to form C-C bonds from aliphatic amides has been a major challenge in Pd catalysis. This study addresses this longstanding problem by using N,N-pym/Boc amides as reagents for cross-coupling and steric tuning of Pd(II)-NHC catalysts. The method is effective for various degrees of aliphatic amide derivatives and the catalyst system is user-friendly. Mechanistic studies support the twist of amide bond and external n(N) -> pi(C=O/Ar)* delocalization as a unified enabling feature of N,N-pym/Boc amides in selective N-C(O) bond activation. This method provides a rare example of Pd-NHC-catalyzed cross-coupling of aliphatic acyl amide electrophiles.

Nickel-Catalyzed Synthesis of an Aryl Nitrile via Aryl Exchange between an Aromatic Amide and a Simple Nitrile

Yang Long et al.

Summary: A nickel-catalyzed synthesis of aryl nitriles via aryl exchange has been developed. The reaction utilizes cheap and easily handled 4-cyanopyridine as the cyanating source, allowing for the efficient conversion of various aromatic amides into aryl nitriles. The reaction exhibits wide substrate scope, good selectivity, and is complementary to traditional methods. Additionally, the mechanism is supported by the determination of two key nickel complexes through X-ray crystallography.

Photoinduced Aerobic Iodoarene-Catalyzed Spirocyclization of N-Oxy-amides to N-Fused Spirolactams**

Loic Habert et al.

Summary: In this study, the aerobic iodoarene catalysis was achieved by using a pyrylium photocatalyst under blue light irradiation, allowing for the use of low catalytic loading of both catalysts under very mild operating conditions. This unprecedented dual organocatalytic system provides a powerful and efficient methodology for spiro-cyclization of amides.

An Efficient Approach for 3,3-Disubstituted Oxindoles Synthesis: Aryl Iodine Catalyzed Intramolecular C-N Bond Oxidative Cross-Coupling

Yang Wang et al.

Summary: In this study, the first intramolecular C-N bond formation of phenylpropanamide derivatives via organocatalytic oxidative reactions was reported, yielding 3,3-disubstituted oxindole derivatives with up to 99% yield. The high efficiency of this reaction is demonstrated by the ability to perform the reaction on a gram scale under transition metal-free mild conditions. Additionally, DFT calculation of the catalytic oxidative transformation pathway was also studied.
