The design of small-molecule prodrugs and activatable phototherapeutics for cancer therapy

Summary: Cancer is a major global health problem, with millions of people diagnosed each year. Current chemotherapy options lack selectivity for cancer cells, are prone to resistance, and have toxicities. Researchers have focused on developing prodrug-based strategies to overcome these limitations. This review discusses recently developed prodrug strategies for cancer therapy and highlights the importance of targeting specific biomarkers in the tumor microenvironment and overexpressed in cancer cells. The advantages and limitations of each approach are also discussed.

Tuning the Solid- and Solution-State Fluorescence of the Iron-Chelator Deferasirox

Summary: Deferasirox, an FDA-approved iron chelator, has been identified as an effective therapeutic agent with fluorescence emission for imaging applications. A library of deferasirox derivatives with different fluorescence emission profiles was designed to overcome overlapping emission wavelengths with other dyes. These derivatives successfully imaged different shapes of bacteria and displayed iron-dependent antibiotic activity. Some derivatives also showed distinct emission features, allowing for multiplex imaging of multiple bacteria.

Production and dispersion of free radicals from transient cavitation Bubbles: An integrated numerical scheme and applications
