Controlling Non-Native Cobalamin Reactivity and Catalysis in the Transcription Factor CarH

Xinhang Yang et al.

Summary: In this study, a variant of the transcription factor CarH, named CarH*, was engineered to catalyze styrene C-H alkylation with improved yields and selectivity compared to cobalamin. Mechanistic studies suggest that the selectivity of CarH* results from its ability to impart a cage effect on radical intermediates, laying the groundwork for the development of B-12-dependent enzymes as catalysts for non-native transformations.

Dual-function enzyme catalysis for enantioselective carbon-nitrogen bond formation

Zhen Liu et al.

Summary: A haem protein has been reported as a dual-function catalyst for inserting a carbene into an N-H bond to form alpha-amino lactones. The engineered cytochrome P450 enzymes demonstrate high activity and enantioselectivity in this enzymatic reaction. Computational studies provide insight into the detailed mechanism and stereocontrol of this new-to-nature reaction.

A Dual Anchoring Strategy for the Directed Evolution of Improved Artificial Transfer Hydrogenases Based on Carbonic Anhydrase

Alina Stein et al.

Summary: Artificial metalloenzymes are hybrid catalysts that anchor a metal cofactor within a host protein to catalyze new-to-nature reactions. In this study, covalent linkage was introduced between the host and guest to improve the localization of a catalyst for enantioselective reduction. Through directed evolution and X-ray analysis, the activity, selectivity, and substrate scope of the catalyst were improved.
