Here, we developed a two-stage isothermal amplification method using strand exchange amplification and hyperbranched rolling circle amplification. This method only requires one DNA polymerase and can be performed in a single closed tube at a constant temperature, offering a promising signal amplification strategy for accurate and rapid pathogen detection in clinical settings.
