Membrane skeleton modulates erythroid proteome remodeling and organelle clearance

Summary: The membrane skeleton plays an important role in modulating proteome remodeling and organelle clearance during final stages of erythropoiesis, with mDia2 acting as a key regulator in this process through polymerization of actin protofilament. Disruption of the skeleton leads to defects in detaching the extruded nucleus and in activating the ESCRT-III complex, resulting in global defects in proteome remodeling, endolysosomal trafficking, and autophagic organelle clearance. Chmp5, regulated by mDia2, is essential for membrane remodeling and autophagosome-lysosome fusion, highlighting a formin-regulated signaling pathway connecting the membrane skeleton to key processes in terminal erythropoiesis.
