Silver-catalyzed cyclopropanation of alkenes with alkynyl N-nosylhydrazones leading to alkynyl cyclopropanes

Summary: A novel method for the stereoselective synthesis of alkynyl cyclopropanes, by the silver-catalyzed alkynylcyclopropanation of alkenes using alkynyl N-nosylhydrazones as alkynyl carbene precursors, is reported. This method provides a straightforward and powerful approach for preparing various alkynyl cyclopropanes in high yield with excellent stereoselectivities. In addition, the practicality of this method was demonstrated by gram-scale synthesis and late-stage modification of bioactive molecules.

Summary: Square-planar cobalt(II) systems play a significant role in the synthesis of (hetero) cyclic compounds through the formation of cobalt(III)-carbene radical intermediates. This study reveals the formation of disubstituted cobalt(III)-carbene radicals and their catalytic application using cobalt(II)-tetraphenylporphyrin complex and iodonium ylides as carbene precursors. The findings highlight the importance of N-enolate formation in the reversible transfer of two carbene moieties.

Base-Promoted Annulative Difluoromethylenation of Enaminones with BrCF2CO2Et toward 2,2-Difluorinated 2,3-Dihydrofurans

Summary: A practical method for the synthesis of 2,2-difluorinated 2,3-dihydrofurans has been developed using enaminones and BrCF2CO2Et as starting materials with Na2CO3 as the catalyst. This method does not require any transition metal reagent and enables the annulative difluoromethylation by partial cleavage of the C=C double bond. Furthermore, treatment with hydrochloric acid in the same reaction system leads to the formation of beta-keto enoic acids via formal enaminone C-N carboxylation.

N-Triftosylhydrazones: A New Chapter for Diazo-Based Carbene Chemistry

Summary: N-sulfonylhydrazones have received significant attention for their ease of preparation, versatile reactivity, high stability, and practicality. N-tosylhydrazones are commonly used, but they require relatively high temperatures for decomposition. Our group has identified N-triftosylhydrazone as an easily decomposable diazo surrogate, allowing for a range of synthetic transformations and expanding the applications of known reactions. In this Account, we report our achievements in using N-triftosylhydrazones in carbene chemistry and categorize the applications based on reaction types. Mechanistic investigations through theoretical calculations provide insights into reaction mechanisms and selectivity.

Construction and transformations of 2,2-difluoro-2,3-dihydrofurans from enaminones and diflurocarbene

Summary: A simple and efficient method for constructing 2-difluoro-2,3-dihydrofurans is reported. The method is metal-free and additive-free, has high functional group tolerance, and uses readily accessible starting materials. Additionally, the synthesized compounds can be easily converted into alpha-amino acids under a reducing atmosphere, and the method can also be used for the synthesis of 2-fluorofurans. This research opens up new avenues for the development of amino acid drugs.

Asymmetric Catalytic Reactions of Donor-Acceptor Cyclopropanes

Summary: The donor-acceptor (D-A) cyclopropanes, due to the synergistic push-pull effect of vicinal electron-donating and electron-withdrawing groups, are recognized as powerful building blocks for generating polyfunctional reactive intermediates. Enantioselective reactions of D-A cyclopropanes provide an efficient approach to enantioenriched acyclic and cyclic compounds. Chiral Lewis/Bronsted acids, transition metals, and organocatalysts have been designed for such transformations, including ring-openings, annulations, and rearrangements.

Construction of C-C Axial Chirality via Asymmetric Carbene Insertion into Arene C-H Bonds
