Straightforward synthesis of quinazolin-4(3H)-ones via visible light-induced condensation cyclization

Wuji Sun et al.

Summary: A green, simple and efficient method for the synthesis of quinazolin-4(3H)-ones has been developed via visible light-induced condensation cyclization. The method utilizes fluorescein as a photocatalyst and TBHP as a reaction promoter without the need for a metal catalyst. The reaction demonstrates high substrate tolerance and yields a variety of desired products in good to excellent yields. This method provides a straightforward strategy for the synthesis of quinazolin-4(3H)-ones.

Discovery of febrifugine with specific anti-Phytophthora oomycete activity isolated from Dichroa febrifuga Lour

Saisai Zhang et al.

Summary: This study found that febrifugine, the main active component of the crude alkaloids extract of D. febrifuga leaves, showed anti-oomycete activity against plant pathogen Phytophthora capsici. Febrifugine altered the morphology of P. infestans hyphae and inhibited zoospore release and germination. In vivo bioassays showed that febrifugine had better protective and curing activities than the commercialized drug metalaxyl. Febrifugine is expected to be a new leading structure for the development of oomyceticides.

Unexpected assembly machinery for 4(3H)-quinazolinone scaffold synthesis

Xi-Wei Chen et al.

Summary: This study reveals a new assembly machinery for 4(3H)-quinazolinone, involving nonribosomal peptide synthase catalysis of tripeptide formation, unusual N-3 sources, and an alpha-ketoglutarate-dependent dioxygenase catalysis of C-N bond oxidative cleavage. The study also uncovers a unique release and tailoring mechanism for nonribosomal peptides and an alternative route for the synthesis of 4(3H)-quinazolinone scaffolds.

Synthesis and in vitro antileishmanial efficacy of novel quinazolinone derivatives

Izak F. Prinsloo et al.

Summary: Current drugs for leishmaniasis treatment have limitations such as high toxicity and emerging resistance, necessitating the development of new, safe, and effective medications. Two new series of synthesized quinazolinone derivatives were assessed for potential antileishmanial properties. While less toxic to mammalian cells compared to halofuginone, the compounds showed modest antileishmanial efficacy, suggesting the need for further structural optimization.

Ruthenium(II)-Catalyzed C-C/C-N Coupling of 2-Arylquinazolinones with Vinylene Carbonate: Access to Fused Quinazolinones

Zhao-Hui Wang et al.

Summary: This work presents a ruthenium(II)-catalyzed C-C/C-N annulation of 2-arylquinazolinones with vinylene carbonate to synthesize fused quinazolinones, showing excellent substrate tolerance and functional-group compatibility. The use of vinylene carbonate as an ethynol surrogate without external oxidants is highlighted, along with preliminary mechanistic studies and a proposed catalytic cycle.

Synthesis of 4(3H)-Quinazolinones by Visible-Light-Induced 2-Benzylaminobenzamides

Xinchang Li et al.

Summary: An environmentally benign protocol of visible-light-induced reaction has been developed for the effective synthesis of 4(3H)-quinazolinones from 2-benzylaminobenzamides. The reaction proceeds well under metal-free, photocatalyst-free and mediator-free conditions, providing the corresponding quinazolinones in good to high yields. Mechanism studies suggest that a radical pathway might be involved in the reaction.
