This review highlights the progress made in the research of small molecule inhibitors targeting Fl-catenin and discusses the future prospects in drug development.

The Casein kinase 1α agonist pyrvinium attenuates Wnt-mediated CK1α degradation via interaction with the E3 ubiquitin ligase component Cereblon

Summary: The Cullin-RING ligase 4 E3 ubiquitin ligase component Cereblon (CRBN) is a target for immunomodulatory drugs that modulate the degradation of neosubstrates required for cancer growth. This study shows that CRBN can also be activated by Wnt ligands and mediates the degradation of certain physiological substrates, including CK1 alpha. A small-molecule CK1 alpha agonist called pyrvinium is found to prevent the interaction between CRBN and CK1 alpha, attenuating CK1 alpha degradation and inhibiting Wnt signaling.
