Quantification of amyloid beta and tau PET without a structural MRI

Susan M. Landau et al.

Summary: This study evaluated several MRI-independent approaches for quantification of positron emission tomography (PET) images. The results showed that MRI-independent methods had high agreement with MRI-dependent quantification in A beta and tau PET images.

Spatial normalization and quantification approaches of PET imaging for neurological disorders

Teng Zhang et al.

Summary: Quantification approaches of PET imaging in living brains have been recommended for clinical diagnosis of neurological disorders. This review compared PET template-based and MRI-aided spatial normalization approaches. The study also discussed ROI-based and voxel-wise quantitative methods, as well as the concerns and clinical applications of these approaches. A recipe for PET spatial normalization and quantification was concluded to improve diagnosis accuracy in clinical practice.

What's the cut-point?: a systematic investigation of tau PET thresholding methods

Alexandra J. Weigand et al.

Summary: This study examined existing studies that derived tau PET cut-points and assessed the validity of different methods of tau PET thresholding. The findings showed variability among studies and provided recommendations for selecting cut-point derivations based on study goals.

Development of a Plasma Biomarker Diagnostic Model Incorporating Ultrasensitive Digital Immunoassay as a Screening Strategy for Alzheimer Disease in a Chinese Population

Xue Wu et al.

Summary: This study evaluated the diagnostic ability of plasma biomarkers for Alzheimer's disease in the Chinese population. The results showed that a simplified diagnostic model combining plasma p-tau181, A beta 42, and clinical features achieved high accuracy and specificity, indicating its potential for large-scale screening in developing countries.

High-Contrast In Vivo Imaging of Tau Pathologies in Alzheimer's and Non-Alzheimer's Disease Tauopathies

Kenji Tagai et al.

Summary: A panel of radiochemicals for in vivo PET imaging of tau pathologies in AD has been developed, but sensitive detection of FTLD tau inclusions remains challenging. The imaging probe PM-PBB3 successfully captures diverse tau deposits in AD and FTLD, with increased binding observed in diseased patients in a subsequent clinical PET study. Visual inspection of F-18-PM-PBB3-PET images aids in individually based identification of diverse clinical phenotypes of FTLD on a neuropathological basis.

Plasma biomarker profiles and the correlation with cognitive function across the clinical spectrum of Alzheimer's disease

Zhenxu Xiao et al.

Summary: Plasma biomarkers in Alzheimer's disease (AD) diagnosis and management are promising, but their profiles and associations with cognition across cognitive stages have been rarely reported. Plasma p-tau181 showed correlations with broader cognitive domains, suggesting its potential as a promising clinical-relevant blood-based biomarker.
