Cortical thickness across the lifespan: Data from 17,075 healthy individuals aged 3-90 years

Sophia Frangou et al.

Summary: The study used data from the ENIGMA Consortium to explore the relationship between age and cortical thickness, finding that most regions peak in cortical thickness during childhood, with a negative association between age and cortical thickness where the slope is steeper before the age of 30 and more gradual afterwards.

Features of the superficial white matter as biomarkers for the detection of Alzheimer's disease and mild cognitive impairment: A diffusion tensor imaging study

Bahare Bigham et al.

Summary: This study applied an automated method to classify patients with Alzheimer's disease (AD) and mild cognitive impairment (MCI) and healthy control (HC) subjects using diffusion tensor imaging (DTI) features in the superficial white matter (SWM). Results showed that the linear kernel of support vector machines (SVM) had the highest accuracy in distinguishing AD and HC groups, followed by the quadratic kernel for MCI and HC groups, and the Gaussian kernel for AD and MCI groups. These features could be a useful tool for assisting in the diagnosis of AD and MCI.

Brain charts for the human lifespan

R. A. I. Bethlehem et al.

Summary: Neuroimaging has become a widely used tool in brain research, but there is currently a lack of reference standards to quantify individual differences over time. In this study, researchers created an open resource that benchmarks brain morphology using a large dataset of MRI scans. The brain charts identified neurodevelopmental milestones and showed high individual stability and robustness to technical and methodological differences.

Surface-based tracking for short association fibre tractography

Dmitri Shastin et al.

Summary: It is estimated that short association fibres (SAF) make up more than half of the total white matter volume in the human brain and play a role in various neurological and psychiatric conditions. However, these fibres remain understudied in neuroimaging research. This study proposes a surface-based approach for mapping short SAF, which produces longer streamlines that connect gyri and provides high cortical coverage. The study also finds that subcortical white matter areas are underrepresented in diffusion-weighted MRI data with lower resolution and weaker diffusion weighting. The reliability of the tractograms is moderate, with low repeatability and high between-subject variability, emphasizing the need for caution when analyzing SAF based on streamline count.

Accelerated decline in white matter microstructure in subsequently impaired older adults and its relationship with cognitive decline

Andrea T. Shafer et al.

Summary: Shafer and Williams et al. found that diffusion tensor imaging can detect faster rates of regionally specific white matter degeneration before symptoms of mild cognitive impairment/dementia are present. This degeneration is associated with faster cognitive decline. Diffusion tensor imaging is a powerful indicator of future risk of developing mild cognitive impairment/dementia.

White matter microstructure across the adult lifespan: A mixed longitudinal and cross-sectional study using advanced diffusion models and brain-age prediction

Dani Beck et al.

Summary: The study reveals that advanced diffusion models provide sensitive measures of age-related microstructural changes in white matter, complementing and extending the conventional DTI contribution. Through linear mixed effects models and machine learning, the research assessed age trajectories in healthy individuals and compared the age prediction accuracy of different diffusion models, showing that advanced models performed similarly to conventional DTI.

Fiber tractography bundle segmentation depends on scanner effects, vendor effects, acquisition resolution, diffusion sampling scheme, diffusion sensitization, and bundle segmentation workflow

Kurt G. Schilling et al.

Summary: This study investigates the impact of various factors on diffusion tractography bundle segmentation, including scan repeats, scanners, vendors, acquisition resolution, diffusion schemes, and diffusion sensitization. The acquisition protocol, particularly resolution, has the largest effect on reproducibility and features variation, followed by vendor and scanner effects. Different segmentation workflows show varying robustness to sources of variation. The choice of bundle segmentation workflows has a bigger impact on the results than other confounding factors.

Tractography dissection variability: What happens when 42 groups dissect 14 white matter bundles on the same dataset?

Kurt G. Schilling et al.

Summary: White matter bundle segmentation using diffusion MRI fiber tractography is a popular method for identifying white matter fiber pathways in human brains, but there is significant variability in segmentation protocols and techniques. A study evaluated the variability among protocols for bundle segmentation and found that even with the same underlying data, the variability across protocols was greater than other sources of variability, indicating a need to reduce heterogeneity for reproducible research and routine clinical use of tractography bundle dissection.

Asymmetric thinning of the cerebral cortex across the adult lifespan is accelerated in Alzheimer's disease

James M. Roe et al.

Summary: Aging and Alzheimer's disease lead to progressive disorganization of the brain, causing a loss of cortical asymmetry that becomes accelerated with age. This study shows that the adaptive asymmetric organization of the heteromodal cortex is dedifferentiated in aging and AD, emphasizing a system-wide impact on brain structure.

Regional Cortical Thickness Predicts Top Cognitive Performance in the Elderly

Elena Nicole Dominguez et al.

Summary: While aging is typically associated with cognitive decline, some individuals can maintain high levels of cognitive performance, with cortical thickness in the cingulate cortex playing a key role. However, utilizing a whole-brain, network-level approach outperformed the localist models, highlighting the importance of considering structural properties across a wide range of areas in understanding cognitive abilities.

Education and age-related differences in cortical thickness and volume across the lifespan

Jason Steffener

Summary: This study found significant moderating effects of age and years of education on gray matter measures in the brain, with the impact of age on gray matter varying across different brain regions and levels of education. Higher education was associated with maintained gray matter in later ages, but also showed a larger decline in gray matter in late life.

Loss and dispersion of superficial white matter in Alzheimer's disease: a diffusion MRI study

Thomas Veale et al.

Summary: Pathological cerebral white matter changes in Alzheimer's disease, particularly in the superficial white matter, were investigated using advanced diffusion MRI techniques in young-onset patients. The results showed decreased neurite density and increased orientation dispersion in the superficial white matter of young-onset Alzheimer's disease individuals, indicating both neurodegenerative and organizational changes. These findings suggest that standard diffusion tensor metrics may not capture the full extent of microstructural alterations in superficial white matter in Alzheimer's disease.

Tract-specific MRI measures explain learning and recall differences in multiple sclerosis

Mia Winter et al.

Summary: Cognitive difficulties are common in multiple sclerosis patients and are closely related to white matter pathology. Studying the relationships between lesions and specific white matter structures can better explain cognitive differences than whole brain imaging. Quantifying the relationships between tract-specific structure and lesions shows promise in elucidating the relationship between neuropathology and cognition in multiple sclerosis.
