C3-Symmetric Ligands in Drug Design: When the Target Controls the Aesthetics of the Drug

Summary: A number of proteins can adopt a homotrimeric spatial conformation, which is crucial for biologic targets in designing C3-symmetric ligands for optimized ligand-target interactions and therapeutic benefits. Both proteins and DNA can benefit from these ligands, even if their conformation does not correspond to a C3-symmetry. The discovery of new homotrimeric targets, such as the SARS-CoV2 spike protein, is expected to increase the use of these ligands in the future. The synthesis of C3-symmetric ligands presents chemical challenges but can result in aesthetically appealing drug candidates.

Functionalization of Covalent Organic Frameworks with DNA via Covalent Modification and the Application to Exosomes Detection

Summary: In this study, we proposed a functionalization strategy for covalent organic frameworks (COFs) using DNA. The DNA-functionalized COFs exhibited good hybridization ability and cargo loading ability. Based on this, an electrochemical biosensor for the detection of exosomes was developed. The functionalized COFs enabled the recognition and capture of exosomes, while the encapsulation of methylene blue in the COFs enhanced the sensitivity of the biosensor.

Room-Temperature Synthesis of 1,3,5-Tri(het)aryl Benzene from Nitroalkenes Using Pd(OAc)2: Complete Mechanistic and Theoretical Studies

Summary: Here, we present a room-temperature catalytic pathway for the synthesis of 1,3,5-tri(het)aryl derivatives from nitroalkenes using simple Pd(OAc)(2). This newly developed method forms C-C bonds in a cascade manner starting from a pallado-Morita-Baylis-Hillman (MBH) type adduct. The broad substrate scopes, functional group tolerance, and different aryl-substituted benzene derivatives make this method more attractive. Furthermore, the isolation of intermediates and DFT studies provide important insights into the mechanism of the catalytic cycle.

Inherently Chiral Cages via Hierarchical Desymmetrization

Summary: A new type of inherently chiral cage structure was obtained through hierarchical desymmetrization, showing rich structural diversity. The enantiopure cages were successfully separated using chiral chromatography and convenient post-synthetic transformations were achieved with retention of enantiomeric purity. The chiral cages consist of three individual chiral cavities, allowing for regio- and enantioselective anion-K binding.

Summary: A new synthetic strategy was reported to synthesize C-3-symmetric molecules with oxepine and benzofuran ring systems, achieved through cyclotrimerization, double bond isomerization, and ring-closing metathesis as key steps. Photophysical property studies using fluorescence spectroscopic data showed strong fluorescence for all compounds except 1 and 2.

Hydrogen-Bonding Controlled Nickel-Catalyzed Regioselective Cyclotrimerization of Terminal Alkynes

Summary: In this study, a hydrogen-bonding controlled nickel-catalyzed regioselective cyclotrimerization of terminal alkynes was reported, yielding 1,3,5-trisubstituted benzenes with high regioselectivities. The method features a low-cost catalyst, mild reaction conditions, and excellent functional group compatibility. The Ni-B(OH)(2) complex generated in situ from NiCl2·DME and tetrahydroxydiboron may act as an active catalyst for this transformation.
