Are Phosphatidylcholine and Lysophosphatidylcholine Body Levels Potentially Reliable Biomarkers in Obesity? A Review of Human Studies

Paula Emilia Nunes Ribeiro Bellot et al.

Summary: Phosphatidylcholines (PCs) are major components of animal membranes and incorporate choline as a headgroup. Lysophosphatidylcholines (LPCs) are derived from the cleavage of PCs and are main components of oxidized low-density lipoproteins (oxLDLs) involved in atherosclerosis. The lipidomic profile changes in obesity, and PCs and LPCs are gaining attention as potential biomarkers. A literature search was performed to clarify this hypothesis, with 21 studies included. Controversial associations between PCs/LPCs and BMI/body fat parameters were identified, along with differences between PCs and LPCs in obesity.

An Overview of Epigenetics in Obesity: The Role of Lifestyle and Therapeutic Interventions

Abeer M. Mahmoud

Summary: Obesity is a global epidemic with genetic predispositions and modern lifestyle characteristics both playing a role. Advances in the study of DNA and its alterations have increased understanding of the epigenetic regulation of energy metabolism in obesity. These reversible epigenetic changes can be targeted for therapeutic interventions.

Metabolic Profiling of Obesity With and Without the Metabolic Syndrome: A Multisample Evaluation

Lars Lind et al.

Summary: This study compared the plasma metabolome in obese subjects with and without metabolic syndrome (MetS) to normal-weight subjects without MetS. The results showed significant differences in metabolite profiles between obese and normal-weight individuals without MetS, with a gradual deterioration in metabolite profile from lean to obese to MetS. The pathways highlighted in this study, such as amino acid synthesis/metabolism and sphingolipid metabolism, may play a role in this metabolic deterioration, with insulin resistance as a potential mediator.

Contribution of specific ceramides to obesity-associated metabolic diseases

Philipp Hammerschmidt et al.

Summary: Ceramides are bioactive membrane sphingolipids that play important regulatory roles in cellular metabolism. Obesity leads to the accumulation of specific ceramides in metabolic tissues, causing metabolic disruptions and the development of cardiometabolic diseases. Recent studies have shown that specific ceramides can affect multiple metabolic pathways, and reducing selected ceramide pools in obese rodents improves metabolic health.

Longitudinal Plasma Lipidome and Risk of Type 2 Diabetes in a Large Sample of American Indians With Normal Fasting Glucose: The Strong Heart Family Study

Guanhong Miao et al.

Summary: The study found that alterations in plasma molecular lipids are closely associated with the risk of type 2 diabetes (T2D) in American Indians, with various lipid species showing significant impact on T2D risk both at baseline and over time. Aberrant lipid profiles were observed in prediabetes, and longitudinal changes in lipidome were significantly linked to changes in continuous measures of T2D. The findings suggest that lipid dysregulation occurs many years before the onset of T2D and may provide new therapeutic targets for early intervention tailored to American Indians.

Gut microbiome and bile acids in obesity-related diseases

Rumei Li et al.

Summary: Dysbiosis is implicated in the etiologies of obesity-related chronic diseases such as type 2 diabetes, NAFLD, and cardiovascular diseases. Bile acids, with their ability to modulate metabolism and immunity through activation of receptors, are recognized as key players in the onset and progression of these diseases. Targeting bile acid signaling pathways or microbiome composition may hold promise in treating obesity-related diseases.

Glycoursodeoxycholic acid ameliorates diet-induced metabolic disorders with inhibiting endoplasmic reticulum stress

Lele Cheng et al.

Summary: Recent studies have shown changes in bile acid metabolite composition in metabolic disorders, with glycoursodeoxycholic acid (GUDCA) being decreased in hyperglycemic patients. GUDCA supplementation alleviated ER stress and apoptosis, stabilizing calcium homeostasis, and ameliorated insulin resistance and hepatic steatosis in HFD-fed mice. This suggests that GUDCA may be a potential treatment option for metabolic disorders by inhibiting ER stress.

Summary: This study reveals marked differences in plasma bile acid profiles between patients with and without T2DM, with higher levels of both primary and secondary bile acids in T2DM patients, as well as specific bile acid species showing significant associations with T2DM status. Further research is needed to elucidate the potential interplay between these differences in plasma bile acid profiles and the pathophysiology of T2DM.
