Predictive high-throughput screening of PEGylated lipids in oligonucleotide-loaded lipid nanoparticles for neuronal gene silencing

Apoorva Sarode et al.

Summary: Lipid nanoparticles (LNPs) have gained attention in nucleic acid delivery field due to the success of mRNA COVID-19 vaccines. PEGylated lipids (PEG-lipids) as one of the constituent lipids on LNP surfaces play a crucial role in defining LNP properties. Research suggests that tuning PEG-lipid parameters can modulate LNP performance. To fully understand LNP behavior, a high-throughput screening (HTS) workflow was developed to prepare, characterize, and assess LNPs loaded with therapeutic antisense oligonucleotide (ASO).

Machine Learning on a Robotic Platform for the Design of Polymer-Protein Hybrids

Matthew J. Tamasi et al.

Summary: This study reports a strategy for designing protein-stabilizing copolymers based on active machine learning, which successfully identified copolymers that preserve or even enhance the activity of three chemically distinct enzymes under thermal denaturing conditions. Although systematic screening results were mixed, active learning appropriately identified unique and effective copolymer chemistries for the stabilization of each enzyme, broadening the capabilities to design fit-for-purpose synthetic copolymers.

Autonomous retrosynthesis of gold nanoparticles via spectral shape matching

Kiran Vaddi et al.

Summary: This paper studies the application of Bayesian optimization in synthesizing complex nanostructures and assemblies. The proposed framework uses shape matching to achieve autonomous retrosynthesis of specific nanoparticles or nano-assemblies. By analyzing the spectra as function spaces and defining a Riemannian metric on them, the framework provides a more accurate shape matching distance compared to the commonly used mean squared error.
