The impact of storage on extracellular vesicles: A systematic study

Stefano Gelibter et al.

Summary: Storage has a significant impact on extracellular vesicles (EVs) samples, leading to a decrease in concentration, reduction in purity, and generation of artefactual particles. It is recommended to process fresh, non-archival samples for EVs analysis, depending on downstream analyses and experimental settings.

Quiescent cancer cells resist T cell attack by forming an immunosuppressive niche

Pilar Baldominos et al.

Summary: This study discovered that in primary breast cancer, tumor cells that resist T cell attack are quiescent and form clusters with reduced immune infiltration. These quiescent cancer cells show superior tumorigenic capacity and higher expression of chemotherapy resistance and stemness genes. Single-cell RNA sequencing revealed differential phenotypes in infiltrating cells based on their intra-tumor location, and uncovered hypoxia-induced immune-suppressive programs. The findings suggest that eliminating quiescent cancer cells holds the promise to counteract immunotherapy resistance and prevent disease recurrence in TNBC.

Regulated IRE1α-dependent decay (RIDD)-mediated reprograming of lipid metabolism in cancer

Aitor Almanza et al.

Summary: The authors discovered that the active form of IRE1 alpha is involved in the reprogramming of lipid metabolism in cancer cells. IRE1 alpha plays a role in cancer progression and regulates lipid metabolism through cleaving mRNAs. The study showed that IRE1 alpha cleaves DGAT2 mRNA, leading to the accumulation of triacylglycerols, and inhibition of IRE1 alpha increases sensitivity to nutritional stress.

S100A9-CXCL12 activation in BRCA1-mutant breast cancer promotes an immunosuppressive microenvironment associated with resistance to immunotherapy

Jianjie Li et al.

Summary: This study explores the mechanism behind the poor response to immune checkpoint blockade in breast cancers caused by BRCA1 deficiency. The authors identify the activation of S100A9-CXCL12 signaling and the accumulation of myeloid-derived suppressor cells as contributing factors. They also demonstrate that the combination of inhibitors targeting S100A9-CXCL12 signaling and alpha PD-1 antibody can effectively suppress the oncogenic actions and improve immunotherapy response.

Extracellular Vesicles: The Landscape in the Progression, Diagnosis, and Treatment of Triple-Negative Breast Cancer

Menglu Dong et al.

Summary: Triple-negative breast cancer (TNBC) is a heterogeneous subtype of breast cancer known for its diverse biological behavior, high aggressiveness, and poor prognosis. Extracellular vesicles (EVs) play a crucial role in long-distance cell-to-cell communication in shaping the tumor microenvironment. EVs released by TNBC cells have potential as valuable biomarkers for diagnosis, monitoring, and treatment evaluation.

Targeting HIF-1α abrogates PD-L1-mediated immune evasion in tumor microenvironment but promotes tolerance in normal tissues

Christopher M. Bailey et al.

Summary: This study found that targeting HIF-1α inhibition can increase the expression of PD-L1 on tumor cells and tumor-infiltrating myeloid cells, activating immune cells and leading to tumor rejection. The HIF-1α inhibitor Echinomycin not only enhances the immunotherapeutic effects of anti-CTLA-4 therapy, but also protects against immune-related adverse events by increasing PD-L1 levels in normal tissues.

S100A8/A9 mediate the reprograming of normal mammary epithelial cells induced by dynamic cell-cell interactions with adjacent breast cancer cells

Seol Hwa Jo et al.

Summary: The direct co-culture of breast cancer cells with non-tumorigenic mammary epithelial cells leads to substantial changes in the molecular and functional characteristics of the mammary epithelial cells, with enhanced proliferation, migration, colony formation, and sphere formation capabilities. Dysregulation of cancer-related pathways is observed, along with upregulation of S100A8 and S100A9, suggesting a potential transition program mediated by these proteins in non-tumorigenic epithelial cells.

Hypoxia in Breast Cancer-Scientific Translation to Therapeutic and Diagnostic Clinical Applications

Ying Zhang et al.

Summary: In breast cancer, intratumoral hypoxia caused by abnormalities in microvasculature is associated with aggressive phenotype, metastasis risk, and resistance to treatments. Hypoxia-inducible factors (HIFs) play a key role in activating gene transcription related to tumor growth, invasion, and maintenance of cancer stem cell properties. Understanding and targeting hypoxia factors in breast cancer may have significant prognostic and predictive value, potentially impacting treatment outcomes.
