Quantification of cell oxygenation in 2D constructs of metallized electrospun polycaprolactone fibers encapsulating human valvular interstitial cells

Summary: The present study introduces a new 2D model for human heart valve in tissue engineering, which incorporates valvular interstitial cells (VIC) encapsulated in gold covered electrospun polycaprolactone (PCL/Au) fibers. The model also serves as an electrochemical sensor for monitoring oxygen levels in cellular media. The biocompatibility of the materials with VIC was evaluated, and electrochemical techniques were used to investigate the effect of cell encapsulation and quantify dissolved oxygen levels. The study provides advancements in the field of heart valve tissue engineering.

Tri-Layered and Gel-Like Nanofibrous Scaffolds with Anisotropic Features for Engineering Heart Valve Leaflets

Summary: In this study, tri-layered gel-like nanofibrous PLGA-PASP scaffolds were successfully fabricated, which featured anisotropic mechanical properties similar to native heart valve tissues and promoted the proliferation and secretion of human aortic valvular interstitial cells. Furthermore, these scaffolds effectively inhibited the osteogenic differentiation of cells. RNA sequencing analysis revealed the significant influence of age on cell behavior.

Summary: In this study, trilayer microfibrous leaflet substrates were produced using the electrospinning technique with two biocompatible and biodegradable polymers, PLLA and PCL. Different concentrations of each polymer were used to create substrates with high mechanical and structural anisotropy. The substrates exhibited cellular adhesion, proliferation, and the production of tissue-engineered constructs when seeded with PVICs. The PLLA substrates showed higher cellular growth and gene expression levels compared to the PCL substrates.
