The auxin-inducible degron 2 (AID2) system enables controlled protein knockdown during embryogenesis and development in Caenorhabditis elegans

Takefumi Negishi et al.

Summary: Targeted protein degradation using the improved AID2 system has been successfully demonstrated in Caenorhabditis elegans, providing rapid and efficient degradation control with lower doses of auxin. This system overcomes the limitations of the previous AID system, including leaky degradation and the difficulty of degrading proteins in embryos. The improved system offers a valuable tool for studying the roles of proteins in embryogenesis and development through temporally controlled protein degradation.

Sterol regulation of developmental and oncogenic Hedgehog signaling

Vikas Daggubati et al.

Summary: This article discusses the role of the Hedgehog (Hh) family of lipid-modified signaling proteins in embryonic tissue patterning and postembryonic tissue homeostasis, as well as the involvement of dysregulated Hh signaling in familial and sporadic cancers. It also explores the binding and modulation of SMO activity by endogenous cholesterol and oxidized cholesterol derivatives. The article proposes models of sterol regulation of SMO and discusses the potential utility of steroidal SMO ligands or inhibitors of enzymes involved in sterol metabolism as cancer therapeutics.

Antisense ribosomal siRNAs inhibit RNA polymerase I-directed transcription in C. elegans

Shimiao Liao et al.

Summary: During the study, it was found that risiRNAs accumulate in RNA exosome mutants in Caenorhabditis elegans and silence pre-rRNAs by directing the association of NRDE proteins with them. The risiRNAs also inhibit transcription elongation of RNA polymerase I and cause mislocalization of exosomes in siRNA suppressor mutants, leading to a novel model of rRNA surveillance combining ribonuclease-mediated RNA degradation and small RNA-directed nucleolar RNAi system.
