New and emerging treatments for inflammatory itch

Stephen Erickson et al.

Summary: Recent therapeutic developments for chronic pruritus have focused on allergic and type 2 inflammatory pathways, targeting histamine, immunoglobulin E, and various cytokines. Advances in neuroimmunology have led to the identification of new therapeutic targets and the rapid development of cutting-edge clinical trials, yet chronic itch remains an area of significant unmet need.

Sex-dependent role of microglia in disulfide high mobility group box 1 protein-mediated mechanical hypersensitivity

Nilesh M. Agalave et al.

Summary: The study explored the differential effects of TLR4-activating HMGB1 on microglia in male and female mice, finding higher cytokine and chemokine expression in males. Only male mice were protected from HMGB1-induced mechanical hypersensitivity with TLR4 ablation in myeloid-derived cells and minocycline treatment. Liquid chromatography-mass spectrometry revealed upregulation of antinociceptive proteins in male mice after minocycline administration, with alpha-1-antitrypsin offering partial protection against HMGB1-induced pain specifically in males.

Central opioid receptors mediate morphine-induced itch and chronic itch via disinhibition

Zilong Wang et al.

Summary: The study demonstrates that intrathecal opioids can induce itching by acting on MOR on spinal inhibitory interneurons, resulting in disinhibition of the spinal itch circuit. In addition, spinal ablation of GRPR(+) neurons can reduce opioid-induced itch, while intrathecal NPY administration can suppress itch response.

Morphine acts on spinal dynorphin neurons to cause itch through disinhibition

Eileen Nguyen et al.

Summary: The study found that morphine-induced itch is caused by neurons rather than mast cells, and spinal dynorphin neurons play an important role in mice. Agonism of the kappa-opioid receptor can alleviate morphine-induced itch, challenging the long-standing use of antihistamines.

IL-23/IL-17A/TRPV1 axis produces mechanical pain via macrophage-sensory neuron crosstalk in female mice

Xin Luo et al.

Summary: The IL-23/IL-17A/TRPV1 axis regulates female-specific mechanical pain, with IL-23 inducing pain in females but impaired in mice lacking IL-23 or IL-23r. IL-23-induced pain is promoted by estrogen and suppressed by androgen.

A new synthetic protectin D1 analog 3-oxa-PD1n-3 DPA reduces neuropathic pain and chronic itch in mice

Jannicke Irina Nesman et al.

Summary: This study focuses on a new analog of protectin D1, 3-oxa-PD1(n-3 DPA), which demonstrated significant alleviation of streptozotocin-induced diabetic neuropathic pain in mouse models as well as itching symptoms in a chronic itch model of cutaneous T-cell lymphoma. These findings highlight the potential of developing new immunoresolvents based on the structure-functions of protectins and their synthetic analogs.

Central opioid receptors mediate morphine-induced itch and chronic itch via disinhibition

Zilong Wang et al.

Summary: The study revealed that intrathecal morphine induces itch by acting on MOR on spinal inhibitory interneurons, leading to disinhibition of spinal itch circuit. The data provide mechanistic insights into the treatment of chronic itch with opioid receptor antagonists like naloxone.

Stepwise crosstalk between aberrant Nf1, Tp53 and Rb signalling pathways induces gliomagenesis in zebrafish

Juanjuan Luo et al.

Summary: The molecular pathogenesis of glioblastoma is critical for human gliomagenesis, with RTK/Ras/PI3K, RB, and TP53 pathways playing key roles. Using transgenic zebrafish with single or multiple gene deletions, researchers induced gliomagenesis and found that nf1 mutation alone had low efficiency, but combined deletions of nf1;tp53 and rb1;tp53 resulted in higher tumor incidences with advanced histology. Bioinformatics analysis revealed alterations in critical pathways like RTK/Ras/PI3K, cell cycle, and focal adhesion, highlighting their importance in gliomagenesis initiation and development. Tumors in zebrafish models showed significant similarities to human gliomas, and temozolomide treatment effectively suppressed gliomagenesis, mimicking responses seen in clinical glioma treatment. This study provides a potential tool for genetically studying gliomagenesis and screening targeted anti-tumor compounds for glioma treatment.
