5-Fluorouracil crystal-incorporated, pH-responsive, and release-modulating PLGA/Eudragit FS hybrid microparticles for local colorectal cancer-targeted chemotherapy

Summary: This study developed 5-FU crystal-incorporated, pH-responsive, and release-modulating poly(D,L-lactide-co-glycolide)/Eudragit FS hybrid microparticles (5FU-EPMPs) for local colorectal cancer (CRC)-targeted chemotherapy. It was found that 5-FU release was prevented in the stomach and small intestine, minimizing systemic absorption. Once reaching the colon, 5-FU was continuously released for over 15 hours, allowing long-term exposure of CRC tissues to sufficient concentrations. The 5FU-EPMPs showed effective inhibition of tumor growth without systemic toxicity in a CRC mouse model.
