Summary: Spatially resolved transcriptomic technologies enable us to study complex biological processes such as mammalian embryogenesis. However, current methods have limitations in resolution, gene capture, and field of view, which hinders their systematic application to large and three-dimensional mid- and late-gestation embryos. In this study, we developed Stereo-seq, a spatially enhanced resolution omics-sequencing method, by combining DNA nanoball-patterned arrays and in situ RNA capture. We used Stereo-seq to generate the mouse organogenesis spatiotemporal transcriptomic atlas, MOSTA, which provides single cell resolution and high sensitivity for mapping the kinetics and directionality of transcriptional variation during mouse organogenesis. By utilizing this atlas, we investigated the molecular basis of spatial cell heterogeneity and cell fate specification in developing tissues like the dorsal midbrain. Our panoramic atlas will facilitate in-depth research into long-standing questions about normal and abnormal mammalian development.

The gene expression in the model resembled that of tumor material. This model allows for research and potential therapy for retinoblastoma in the human retina.

Summary: Mutations in PRPF31, a core protein of the spliceosomal tri-snRNP complex, cause autosomal-dominant retinitis pigmentosa (adRP). This study used iPSC technology to generate retinal organoids and RPE models from PRPF31-adRP patients, and found that RNA splicing, autophagy and lysosome, and visual cycle pathways were significantly affected. The accumulation of cytoplasmic aggregates and impaired waste disposal mechanisms were also observed in patient-derived cells. Activation of the autophagy pathway reduced the aggregates and improved cell survival.

Development of human retinal organoid models for bisphenol toxicity assessment

Summary: Bisphenols, such as Bisphenol A (BPA), Tetrabromobisphenol A (TBBPA), and Tetrabromobisphenol S (TBBPS), have been widely used and detected in the environment. While the toxic effects of bisphenols on human embryonic development have been studied, their effects on retinal development are still unknown. This study used human embryonic stem cells to create retinal organoids and found that bisphenols interfered with retinal development processes and the Cytokine-cytokine receptor interaction pathway. Different bisphenols also exhibited different toxic effects on neural retina development, depending on the developmental stage. This study provides new insights into the retinotoxicity after prenatal bisphenol exposure.
