Synthesis, kinetic studies and in-silico investigations of novel quinolinyl-iminothiazolines as alkaline phosphatase inhibitors

A series of novel quinolinyl iminothiazolines was synthesized and evaluated for alkaline phosphatase inhibition potential. Compound N-benzamide quinolinyl iminothiazoline (6g) exhibited the maximum inhibitory effect and can be suggested for further testing.

Discovery of chalcone derivatives as potential α-glucosidase and cholinesterase inhibitors: Effect of hyperglycemia in paving a path to dementia

Summary: A series of fascinating chalcone derivatives 1C-14C were designed and synthesized by Claisen-Schmidt condensation, and their inhibitory activities against alpha-glucosidase and cholinesterase were screened. The target compounds were evaluated for their antihyperglycemic and neuroprotective activities using in vivo mechanism-based assays. Some of these compounds showed significant antidiabetic and neuroprotective activities and demonstrated a strong relationship between their structures and selected properties. They could potentially be used for age-related problems and neurodegenerative disorders.

Exploring thiazole-linked thioureas using alkaline phosphatase assay, biochemical evaluation, computational analysis and structure-activity relationship (SAR) studies

Summary: This study synthesized a new group of thiazole-linked thioureas with aromatic and aliphatic side chains using a one-pot three-component strategy. The synthesized compounds were evaluated for their alkaline phosphatase inhibitory activity, and their chemical structures were identified by NMR. Density functional theory and molecular docking studies were conducted to understand the reactivity and binding behavior of the compounds. The results showed that the synthesized compounds are potent alkaline phosphatase inhibitors, with 4c and 4g being the most active compounds. These derivatives have the potential to be used for the synthesis of more potent alkaline phosphatase inhibitors for the treatment of different types of cancer.

Potent Alkaline Phosphatase Inhibitors, Pyrazolo-Oxothiazolidines: Synthesis, Biological Evaluation, Molecular Docking, and Kinetic Studies

Summary: A series of pyrazolo-oxothiazolidine derivatives were synthesized and evaluated for their inhibitory activity against alkaline phosphatase. The most potent compound, 7g, demonstrated strong inhibition and non-competitive binding with the enzyme. These compounds also exhibited antioxidant activity and showed no toxicity to human osteosarcoma cells at a certain concentration. Computational research confirmed their good binding affinity with alkaline phosphatase, suggesting their potential as selective inhibitors.

Molecular docking and quantitative structure-activity relationship studies were performed to understand the binding interactions and establish a correlation between the compound structures and their properties. These findings suggest that these novel compounds could be potential leads for the development of new drugs with anti-hyperglycemic effects.

Exploring Novel Pyridine Carboxamide Derivatives as Urease Inhibitors: Synthesis, Molecular Docking, Kinetic Studies and ADME Profile

Summary: The rapid development of resistance by ureolytic bacteria, such as those associated with gastric and duodenal cancer, has created a need for a new therapy with anti-urease activity. A series of pyridine carboxamide and carbothioamide derivatives were synthesized and investigated for their inhibitory action against urease. Two derivatives, 5-chloropyridine-2-yl-methylene hydrazine carbothioamide (Rx-6) and pyridine 2-yl-methylene hydrazine carboxamide (Rx-7), exhibited significant activity. Molecular docking and kinetic studies were conducted to understand the binding mode and interaction of the most potent inhibitors with urease. ADME profile showed that all synthesized molecules had oral bioavailability and high GI absorption.
