Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Metastatic triple negative breast cancer adapts its metabolism to destination tissues while retaining key metabolic signatures

Fariba Roshanzamir et al.

Summary: This study utilized transcriptomic data and metabolic network analyses to investigate the adaptation of metastatic tumors to the local metabolic environment. The findings suggest that metastatic tumors adopt a metabolic signature similar to their destination's primary tumors, but the extent of adaptation varies across different organs. Metastatic tumors also retain metabolic signatures associated with TNBC.

Triple negative breast cancer: Pitfalls and progress

Paola Zagami et al.

Summary: Triple negative breast cancer is a subtype of breast cancer characterized by the lack of hormone receptors and HER2 expression. It has a higher incidence among Black and pre-menopausal women and a poorer prognosis. Currently, there is a lack of targeted therapy for TNBC, but early outcomes have improved with intensified treatments and immunotherapy. Future efforts are needed to identify targetable abnormalities, prevent immune evasion, and provide more social-economic support.

SIRT6 in Aging, Metabolism, Inflammation and Cardiovascular Diseases

Zhenyang Guo et al.

Summary: SIRT6, as an important NAD(+)-dependent enzyme, plays multiple roles in cellular signaling pathways and is closely associated with various physiological and pathological processes such as aging, metabolism, inflammation, and cardiovascular diseases. This article provides a detailed description of the specific substrates and molecular mechanisms of SIRT6, and reviews the recent advances in SIRT6 modulators as potential therapeutic agents for SIRT6-mediated diseases.

Breast Tumor Kinase (Brk/PTK6) Mediates Advanced Cancer Phenotypes via SH2-Domain Dependent Activation of RhoA and Aryl Hydrocarbon Receptor (AhR) Signaling

Amy R. Dwyer et al.

Summary: Research shows that in triple-negative breast cancer, the PTK6-driven signaling pathway mediates cell migration via RhoA and AhR signaling pathways, and plays a crucial role in lung metastasis.
