Martini 3 Coarse-Grained Force Field: Small Molecules

Riccardo Alessandri et al.

Summary: The recent re-parametrization of the Martini coarse-grained force field, Martini 3, has improved the accuracy of the model in predicting molecular packing and interactions in molecular dynamics simulations. The use of higher resolution coarse-grained particles to describe ring-like structures in small molecules has resulted in excellent partitioning behavior and solvent properties, as well as capturing miscibility trends between different bulk phases.

Cryo-EM Structure of Mechanosensitive Channel YnaI Using SMA2000: Challenges and Opportunities

Claudio Catalano et al.

Summary: Mechanosensitive channels respond to mechanical forces on the cell membrane, but high-resolution structural information is needed to understand gating mechanisms. Protein-lipid interactions are crucial for these channels, but detergents cannot maintain native lipid environment. Detergent-free systems are emerging as alternatives, but there are limitations in resolving complete structures.

Detergent-free systems for structural studies of membrane proteins

Youzhong Guo

Summary: Membrane protein structural biology has traditionally relied on detergent-based methods, but detergent-free methods have recently emerged as promising alternatives. Among the current detergent-free technologies, the NCMN system shows improved maintenance of native cell membrane lipids and is suitable for high-resolution structural analysis of membrane proteins. Continued development and optimization of new membrane-active polymers will further advance these detergent-free systems.

Biological insights from SMA-extracted proteins

Lucas Unger et al.

Summary: The use of styrene maleic acid (SMA) for extracting membrane proteins has provided rich structural data and insights into protein-lipid interactions. Despite limitations such as sensitivity to low pH, SMA's key advantage lies in maintaining the protein's lipid bilayer, allowing researchers to study proteins in a near-native environment.

Structural biology of endogenous membrane protein assemblies in native nanodiscs

Chanelle J. Brown et al.

Summary: The development of amphiphilic copolymers has allowed the solubilization of integral membrane proteins into native nanodiscs, providing insights into their structures and functions. This breakthrough has established biological membrane:protein assemblies as fundamental units of cellular membranes, and various methods including cryo-EM and NMR spectroscopy have been utilized for the characterization of transmembrane proteins within these nanodiscs.
