NLRP3 inflammasome in neurodegenerative disease

Summary: Neurodegenerative diseases are characterized by dysregulated neuro-glial microenvironment and chronic inflammation, which can lead to progressive neuronal loss. The NLRP3 inflammasome has gained interest as a potential target for anti-inflammatory treatments due to its involvement in recognizing pathologic forms of amyloid, tau, and alpha-synuclein. However, there are still gaps in our understanding of how intracellular modifiers and resolution mechanisms regulate inflammasome activation. Further analysis of inflammasome-driven pyroptosis and inflammation in neurodegenerative diseases is important for developing effective therapeutics and biomarkers.

The synthesis involved directed C-H oxidation, Wagner-Meerwein-type double methyl migration, Meinwald rearrangement, and double-bond isomerization/spiroketal formation cascade. The proposed biosynthetic speculation was modified by this synthetic sequence, which also served as a platform for the synthesis of other lanostanes with migrating methyl groups.

Cryptotanshinone specifically suppresses NLRP3 inflammasome activation and protects against inflammasome-mediated diseases

Summary: Cryptotanshinone is a specific inhibitor of the NLRP3 inflammasome, blocking its activation by inhibiting upstream signals such as Ca2+ signaling and mitochondrial reactive oxygen species induction. In mouse models, cryptotanshinone attenuates caspase-1 activation and IL-1 beta secretion in NLRP3 inflammasome-mediated diseases.
