Generation and Use of Glycosyl Radicals under Acidic Conditions: Glycosyl Sulfinates as Precursors

Summary: We present a method for generating glycosyl radicals under highly acidic conditions. This method utilizes glycosyl sulfinates as radical precursors, which are stable solids and can be easily prepared from commercial starting materials. The method allows for direct installation of glycosyl units onto pyridine rings through the Minisci reaction. We showcase the utility of the method in the late-stage modification of complex drug molecules, including camptothecin. Experimental studies provide insight into the reaction mechanism.

Carbohydrate-DNA Conjugation Enabled by Glycosyl Radicals Generated from Glycosyl Sulfinates

Summary: Here, a method for synthesizing carbohydrate-DNA conjugates by radical addition is reported. This method utilizes readily available, bench-stable, and unprotected glycosyl sulfinates as precursors to glycosyl radicals. The reaction, which is redox neutral, proceeds under mild and simple conditions and allows for a broad substrate scope. A small library of carbohydrate-DNA conjugates was successfully prepared.

Carbon-Centered Radicals in Protein Manipulation

Summary: Methods to directly modify proteins post-translationally have potential for creating and studying protein post-translational modifications. Recent developments in single electron/radical chemistry have allowed for site-specific editing of proteins. This review gives an overview of current methods that install such edits, focusing on radical C-C bond formation and its compatibility with proteins and cells, as well as the novel applications afforded by these methods. Despite technical challenges, radical C-C bond formation on proteins is a promising and rapidly growing field in chemical biology.

Summary: This review article summarizes selected literature from 2000 to 2022 on the synthetic development of C-glycosylation involving the generation of glycosyl radical as a crucial intermediate. The methods for controlling the generation of glycosyl radical are categorized based on the key source of the initiator, and the stereochemical outcome of the resultant C-glycosides is discussed. The synthesis of challenging C-linked building blocks via the generation of glycosyl radical provides a more facile approach for the development of drugs and inhibitors in the field of biological and medicinal chemistry.
