Gold(I)-Catalyzed Benzylic C(sp3)-H Functionalizations: Divergent Synthesis of Indole[a]- and [b]-Fused Polycycles

Summary: In this study, phenyl azides substituted by an (alkylphenyl)ethynyl group were found to facilitate benzylic sp(3)(C-H) functionalization under the presence of a JohnPhosAu catalyst, resulting in the synthesis of indole-fused tetra- and pentacycles through divergent N- or C-cyclization. The chemoselectivity of the reaction was found to be influenced by the counter-anion, the electron density of the alpha-imino gold(I) carbene, and the alkyl groups stabilizing the benzylic carbocation.

Direct Access to 9/6-Fused Cycles via Sequential Hydride Shift Mediated Double C(sp3)-H Bond Functionalization

A Synthesis of Alstonlarsine A via Alstolucines B and F Demonstrates the Chemical Feasibility of a Proposed Biogenesis

Summary: We propose a new biogenetic explanation for the origin of the complex alkaloid alstonlarsine A, involving the rearrangement of the Strychnos alkaloids alstolucines B and F. Additionally, we provide evidence for the chemical feasibility of this proposal through a sequence of N-methylation, beta-elimination, and a cascade 1,7-hydride shift/Mannich cyclization. Notably, this proposal involves the tert-amino effect, which serves as a hydride-shift-based internal redox trigger for the Mannich cyclization. The stereochemical outcome of the cascade reaction is likely influenced by conformational preferences during the hydride shift.

Summary: Direct functionalization of inert C-H bonds is a powerful strategy for constructing complex structures. However, existing methods face challenges in meeting the requirements of modern green chemistry. Over the past decades, the use of hydride shift activation in the functionalization of inert C(sp(3))-H bonds has gained attention. This review summarizes the significant advances in cascade [1,5]-hydride shift/intramolecular C(sp(3))-H functionalization, focusing on the synthesis of spiro-tetrahydroquinoline derivatives containing various pharmaceutical units. Current limitations and future directions in this field are also discussed.

Deuterium kinetic isotope effects as redox mechanistic criterions

Summary: This article focuses on deuterium kinetic isotope effects (KIEs) as criteria for elucidating redox mechanisms, including proton transfer, hydrogen transfer, and hydride transfer reactions. Different reaction pathways and mechanisms are discussed based on the redox properties of donors and acceptors, exhibiting unique KIEs. Temperature dependency of KIEs in aldehyde deformylation reactions distinguishes between two reaction pathways, shedding light on the mechanisms involved.

Synthesis of spiro[imidazole-4,3′-quinolin]ones from [2-(dimethylamino)benzylidene]-2-(methylsulfanyl)imidazolones

Summary: Treatment of 5-[2-(dimethylamino)benzylidene]-3-methyl-2-(methylsulfanyl)-3,5-dihydro-4H-imidazol-4-ones with catalytic scandium triflate resulted in [1,5]-hydride shift followed by cyclization, yielding 1,1'-dimethyl-2-(methylsulfanyl)-1',4'-dihydro-2'Hspiro[imidazole-4,3'-quinolin]-5(1H)-ones. Compounds with various substituents in the benzylidene moiety were successfully synthesized in good yields.
