TRAPPC1 is essential for the maintenance and differentiation of common myeloid progenitors in mice

Summary: This study reveals the essential role of TRAPPC1 in the maintenance and differentiation of CMPs and highlights the significance of protein processing and trafficking processes in myeloid cell development. TRAPPC1 deficiency causes severe monocyte and neutrophil defects, accompanied by a selective decrease in CMPs and subsequent cell subsets in bone marrow. TRAPPC1-deleted CMPs differentiate poorly into monocytes and neutrophils in vivo and in vitro, in addition to exhibiting enhanced endoplasmic reticulum stress and apoptosis via a Ca2+-mitochondria-dependent pathway.

Trappc1 deficiency impairs thymic epithelial cell development by breaking endoplasmic reticulum homeostasis

Summary: This study investigates the role of transport protein particle complexes (Trappcs) in the development and maturation of thymic epithelial cells (TECs). The researchers find that Trappc1 deficiency leads to severe thymic atrophy, decreased TEC cell numbers, and impaired TEC maturation. Additionally, TEC-specific Trappc1 deletion results in poor thymic T cell output and increased activation of memory T cells, contributing to spontaneous autoimmune disorders. Molecular studies reveal that reduced endoplasmic reticulum and Golgi apparatus, enhanced unfolded protein response, apoptosis, and reactive oxygen species generation are involved in the reduction of Trappc1-deleted TECs.

Allosteric inhibition of PPM1D serine/threonine phosphatase via an altered conformational state
