An alternative, zeaxanthin epoxidase-independent abscisic acid biosynthetic pathway in plants

Summary: This study discovered a ABA biosynthetic pathway that is independent of ABA1 and identified the intermediates of this pathway as apocarotenoids. These apocarotenoids are successively converted into ABA in both Arabidopsis and rice, exerting ABA biological functions such as maintaining seed dormancy and inducing the expression of ABA-responsive genes. Furthermore, these apocarotenoids also exhibit ABA-independent regulatory activities.

By obtaining axenic lines through zygospores treatment and antibiotic cocktail, and utilizing PEG-mediated protoplast transformation and particle bombardment, successful transient transgene expression was achieved. Moreover, a higher promoter activity using the 5' -flanking region of the MrACT1 gene fused with green fluorescent protein was observed in comparison with heterologous promoters.

Summary: Phylogenomic analyses of RLKs across plants revealed their presence in streptophytes, some chlorophytes, one prasinodermophyte, and one glaucophyte, but not in rhodophytes. The copy number of RLKs in streptophytes increased drastically after a certain point in evolution. Charophyte RLKs are diverse and seed the major diversity of land plant RLKs, suggesting modular evolution through domain gains or losses. Positive selection in charophyte RLK groups indicates potential functions in host-microbe interaction.

Unexpected cryptic species among streptophyte algae most distant to land plants

A clade consisting of Mesostigmatophyceae, Spirotaenia spp., and Chlorokybophyceae is most divergent from land plants. Within Chlorokybus, deep genetic structures were uncovered, leading to the description of four new species. Gene expression differences among Chlorokybus species suggest constitutive variability that may impact their response to environmental factors.

Despite broad similarities between CAM in Isoetes and terrestrial angiosperms, several key differences are identified, including the recruitment of 'bacterial-type' PEPC and diverged circadian control of key CAM pathway genes in Isoetes.
