Suppression of fibrin(ogen)-driven pathologies in disease models through controlled knockdown by lipid nanoparticle delivery of siRNA

Lih Jiin Juang et al.

Summary: Fibrinogen plays a pathologic role in multiple diseases, but therapeutic strategies to regulate its concentration are lacking. This study developed a strategy using lipid nanoparticle-encapsulated small interfering RNA to modulate fibrinogen expression, effectively reducing fibrinogen levels.

Liver injury in non-alcoholic fatty liver disease is associated with urea cycle enzyme dysregulation

Rocio Gallego-Duran et al.

Summary: This study aimed to evaluate changes in urea cycle enzymes in NAFLD patients and animal models, and found that liver fibrosis and steatosis were associated with a reduction in both gene and protein expression patterns of mitochondrial urea cycle enzymes.

Defining IL-6 levels in healthy individuals: A meta-analysis

Elias A. Said et al.

Summary: This meta-analysis is the first to report IL-6 levels in the blood of healthy donors based on a large number of studies, with a pooled estimate of 5.186 pg/ml. Results showed that IL-6 values increased by 0.05 pg/ml for every 1 year increase in age. Despite heterogeneity, differences in IL-6 values were within a few pg/ml range, possibly due to varying conditions impacting IL-6 production in the healthy population.

Clinical phenotype, fibrinogen supplementation, and health-related quality of life in patients with afibrinogenemia

Alessandro Casini et al.

Summary: The study highlights the severe bleeding and thrombotic phenotype of afibrinogenemia and its impact on health-related quality of life. Younger age, residence in Asia or Africa, and a previous thrombotic event are statistically correlated with a worse HRQoL. Further research is needed to determine the optimal strategy for fibrinogen supplementation.

The regulation of Hypoxia-Inducible Factor-1 (HIF-1alpha) expression by Protein Disulfide Isomerase (PDI)

Yukino Kobayashi et al.

Summary: The study reveals that PDI regulates HIF-1α by modulating its redox state for lysosome-dependent degradation, making it a novel factor in controlling HIF-1α.
