Lignin peroxidase-catalyzed direct oxidation of trace organic pollutants through a long-range electron transfer mechanism: Using propranolol as an example

Summary: In this study, lignin peroxidase (LiP) was extracted and used for the in vitro degradation of the persistent compound propranolol (PPN). The results showed that LiP was able to degrade a high percentage of PPN under specific conditions. The study also revealed two different electron-transfer routes in the LiP degradation mechanism and deduced possible degradation pathways through mass-spectrometry analysis. The results demonstrated that LiP has a remarkable capacity for removing various recalcitrant pollutants in environmental remediation.

Multifunctional P450 Monooxygenase CftA Diversifies the Clifednamide Pool through Tandem C-H Bond Activations

Summary: This study provides detailed insights into the biosynthetic mechanism of polycyclic tetramate macrolactams (PTMs), particularly the role of P450 monooxygenases encoded in PTM biosynthetic gene clusters (BGCs). The multifunctional P450 monooxygenase CftA is shown to catalyze the conversion of ikarugamycin to clifednamide A or clifednamide C through C-H bond activation. The complete structure of clifednamide C is also revealed. These findings expand our understanding of PTM biosynthesis and the customization of PTM natural products.

Different Geometric Requirements for Cytochrome P450-Catalyzed Aliphatic Versus Aromatic Hydroxylation Results in Chemoselective Oxidation

Molecular dynamics simulations suggest that the different geometrical requirements for efficient aromatic oxidation versus aliphatic C-H bond hydroxylation by cytochrome P450 enzymes result in chemoselectivity.

An Altered Heme Environment in an Engineered Cytochrome P450 Enzyme Enables the Switch from Monooxygenase to Peroxygenase Activity

Summary: In this study, it was found that mutating a highly conserved threonine residue in CYP199A4 to glutamate can convert it to a peroxygenase and improve its peroxygenase activity. The crystal structure of the mutant demonstrated a change in spin state upon substrate binding, which was linked to the modification of the oxygen-binding groove and the retention of the iron-bound aqua ligand. The detailed characterization of this engineered heme peroxygenase will aid in the development of new methods for the selective C-H bond activation using heme enzymes.

Oxidation of dobutamine and dopamine by horseradish peroxidase

Exploring the Potential of Cytochrome P450 CYP109B1 Catalyzed Regio-and Stereoselective Steroid Hydroxylation
