Enantioselective Synthesis of N-N Atropisomers by Palladium-Catalyzed C-H Functionalization of Pyrroles

Summary: In this study, a palladium-catalyzed enantioselective C-H activation method for pyrroles was reported, which successfully synthesized structurally diverse indole-pyrrole atropisomers with a chiral N-N axis, showing high yields and enantioselectivities. Furthermore, the kinetic resolution of trisubstituted N-N heterobiaryls with more sterically demanding substituents was also achieved. This versatile method enables the rapid and selective functionalization of pyrroles, facilitating the synthesis of valuable and complex N-N atropisomers.

Chiral Acid-Catalyzed Atroposelective Indolization Enables Access to 1,1′-Indole-Pyrroles and Bisindoles Bearing a Chiral N-N Axis

Summary: We developed a highly selective method for synthesizing 1,1'-biheteroaryls bearing a chiral N-N axis. By using chiral phosphoric acid and InBr3 as cooperative catalysts, 2,3-diketoesters reacted with 1,3-dione-derived enamines to construct 1,1'-pyrrole-indoles with high enantioselectivity, up to 92% yield, and up to 94% enantiomeric excess (ee), or bisindoles with up to 92% ee. Derivatizations of these compounds resulted in diverse functionalized N-N linked axially chiral biheteroaryls.

Asymmetric synthesis of N-N axially chiral compounds via organocatalytic atroposelective N-acylation

Summary: This study reported the first atroposelective N-acylation reaction for the direct catalytic synthesis of optically active atropisomeric quinazolinone derivatives. The reaction features mild conditions, broad substrate scope, high yields, and very good enantioselectivities. The synthetic utility of the protocol was supported by a large scale reaction, transformation of the product, and the utilization of the product as an acylation kinetic resolution reagent, with DFT calculations providing convincing evidence for stereoselection interpretation.

Rhodium(III)-Catalyzed Enantioselective C-H Activation/Annulation of Ferrocenecarboxamides with Internal Alkynes

Summary: In this study, an enantioselective C-H activation/annulation reaction catalyzed by CpRh(III) was reported for the efficient synthesis of planar chiral ferrocene-fused heterocyclic molecules. Cu(I)-Catalyzed Asymmetric Arylation of Pyrroles withDiaryliodonium Salts toward the Synthesis of N-N Atropisomers

Summary: In this study, we demonstrate that copper(I) catalysis using a bis(phosphine) dioxide ligand can efficiently catalyze the desymmetric C-H arylation of prochiral bipyrroles with excellent enantioselectivities. More than 50 nitrogen-nitrogen atropisomers were achieved in good to excellent yields. The mild reaction conditions and good functional group compatibility on arenes and diaryliodonium salts make this method highly versatile.

Several strategies have been developed based on the presence or absence of symmetry in the starting material and the occurrence of racemization or dynamicization events, including desymmetrization, classical kinetic resolution (KR), dynamic kinetic resolution (DKR), and dynamic kinetic asymmetric transformation (DYKAT). Seminal contributions and selected examples are discussed to highlight the potential of these synthetic tools.
