Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

PCK2 opposes mitochondrial respiration and maintains the redox balance in starved lung cancer cells

Gabriele Bluemel et al.

Summary: PCK2 acts as a regulator of mitochondrial respiration and maintains redox balance in nutrient-deprived cancer cells. Inhibition of PCK2 significantly reduces colony formation of lung cancer cells under starvation, showing its potential as a therapeutic target.

O-GlcNAc modified-TIP60/KAT5 is required for PCK1 deficiency-induced HCC metastasis

Rui Liu et al.

Summary: Aberrant glucose metabolism and elevated O-linked beta-N-acetylglucosamine modification are common in hepatocellular carcinoma (HCC), with loss of phosphoenolpyruvate carboxykinase 1 (PCK1) leading to increased protein O-GlcNAcylation and promoting HCC metastasis. Through gain- and loss-of-function studies, it was demonstrated that PCK1 suppresses HCC metastasis by regulating KAT5 O-GlcNAcylation and epigenetically activating TWIST1 and c-Myc to promote epithelial-mesenchymal transition (EMT) in HCC. Targeting HBP-mediated O-GlcNAcylation of KAT5 inhibits lung metastasis of HCC in hepatospecific Pck1-deletion mice, highlighting the link between metabolic enzyme deficiency and epigenetic regulation in HCC metastasis.

Gluconeogenic enzyme PCK1 deficiency promotes CHK2 O-GlcNAcylation and hepatocellular carcinoma growth upon glucose deprivation

Jin Xiang et al.

Summary: Cancer cells with elevated HBP activity and O-GlcNAcylation are associated with rapid tumor growth. PCK1 knockout leads to increased O-GlcNAcylation under low-glucose conditions, promoting HCC development. Inhibitors targeting HBP-mediated O-GlcNAcylation could be potential therapeutic targets for HCC treatment.

The S100 calcium-binding protein A11 promotes hepatic steatosis through RAGE-mediated AKT-mTOR signaling

Fei Teng et al.

Summary: The study demonstrated that S100A11 is upregulated in NAFLD patients and overexpression of S100A11 significantly increases liver steatosis, body weight, and AST levels. Mechanistically, S100A11 acts as a positive regulator of the AKT/mTOR signaling pathway to induce lipid synthesis and aggravate lipid deposition. Targeting S100A11 may be a potential therapeutic approach for NAFLD.

Phosphoenolpyruvate carboxykinase in cell metabolism: Roles and mechanisms beyond gluconeogenesis

Shuo Yu et al.

Summary: PCK enzyme plays important roles beyond gluconeogenesis, including in the development of obesity, cardiac hypertrophy, stroke, and cancer. The expression of PCK is precisely controlled by a complex regulatory network involving epigenetic regulation, transcription regulation, and posttranscription regulation. Understanding the regulation of PCK may lead to the development of strategies to treat metabolic dysfunction.

Cul4A-DDB1-mediated monoubiquitination of phosphoglycerate dehydrogenase promotes colorectal cancer metastasis via increased S-adenosylmethionine

Yajuan Zhang et al.

Summary: This study revealed a new role of SAM in promoting tumor metastasis through PHGDH activity regulation by monoubiquitination in colorectal cancer cells. The increased levels of SAM led to upregulation of cell adhesion genes, promoting tumor cell migration and CRC metastasis. Interestingly, SAM levels correlated with metastatic recurrence in CRC patients.
