p21 induces a senescence program and skeletal muscle dysfunction

Summary: Recent research has found associations between elevated levels of p21, the accumulation of senescent cells, and skeletal muscle dysfunction in mice and humans. By using a mouse model with p21 overexpression, the study investigated the role of p21 in cellular senescence and pathological features in skeletal muscle. The findings showed that p21 induced several characteristic features of cellular senescence in skeletal muscle, including altered gene expression, DNA damage, mitochondrial dysfunction, and the senescence-associated secretory phenotype. Additionally, the p21OE mice exhibited skeletal muscle pathology, such as atrophy, fibrosis, and impaired physical function, highlighting p21 as a driver of cellular senescence and a new source of skeletal muscle loss and dysfunction.

Advancing our understanding of skeletal muscle across the lifecourse: Protocol for the MASS_Lifecourse study and characteristics of the first 80 participants

However, there has been limited investigation of sarcopenia using experimental medicine techniques. The analysis of the characteristics of the first 80 participants showed that their grip strength was similar to the general population, and the findings from this study should contribute to improvements in the diagnosis, treatment, and prevention of sarcopenia.

Characterization of cellular senescence in aging skeletal muscle

Summary: This study comprehensively assesses cellular senescence in skeletal muscle of young and old mice using single-cell and bulk RNA sequencing, as well as imaging methods. The results demonstrate that a subpopulation of fibroadipogenic progenitors in old mice highly express p16(Ink4a) and multiple senescence-related genes, accompanied by DNA damage and chromatin reorganization. A senescence phenotype is also observed in isolated myofibers, governed by p21(Cip1). Treatment with a senotherapeutic intervention improves age-related molecular and morphological changes and enhances skeletal muscle strength in old mice. Furthermore, the senescence phenotype is conserved in skeletal muscle from older humans.
