The Application of Biomaterials in Spinal Cord Injury

Summary: The spinal cord and brain are vital to the central nervous system (CNS), but spinal cord injury (SCI) caused by external forces is challenging to treat and results in loss of motor, sensory, and autonomic functions. The incidence of SCI is increasing globally, emphasizing the urgent need for treatments. Various approaches, such as surgery, drug therapy, stem cell transplantation, regenerative medicine, and rehabilitation therapy, have been developed. Among them, biomaterials using tissue engineering and bioscaffolds show promise for transporting cells or drugs to injured sites. However, clinical evidence for the treatment of SCI with biomaterials remains limited. This review provides inspiration and guidance for future studies and applications of biomaterials in SCI treatment.

Current Concepts of Biomaterial Scaffolds and Regenerative Therapy for Spinal Cord Injury

Current management approaches focus on symptomatic treatment, lacking effective therapeutic strategies. However, recent advancements in regenerative medicine have shown promise in preclinical and clinical studies. The utilization of biomaterial scaffolds, combined with stem cell therapy or other regenerative therapies, holds great potential in accelerating the development of novel therapeutic strategies for SCI. This review summarizes the latest insights from preclinical and clinical studies utilizing biomaterial scaffolds for regenerative therapy and provides a comprehensive overview of the biomaterial strategies employed.

Intracutaneous Transplantation of Islets Within a Biodegradable Temporizing Matrix as an Alternative Site for Islet Transplantation

Summary: A clinically proven biodegradable temporizing matrix (BTM) has been found to facilitate the formation of a well-vascularized intracutaneous neodermis within the skin, which improves the survival and functionality of transplanted islet cells. The experiments conducted on mice and pigs showed that BTM does not harm the viability and function of islet cells, and it can maintain the normal structure and function of the islets. BTM has the potential to become an alternative site for islet transplantation.

Composite Fibrin and Carbon Microfibre Implant to Modulate Postraumatic Inflammation after Spinal Cord Injury

Summary: Poor functional recovery after spinal cord injury (SCI) has prompted the development of novel strategies. Our recent study demonstrated the immunomodulatory and pro-regenerative effects of bio-functionalized carbon microfibres (MFs) in a rat model of SCI. To enhance tissue repair and ease MF implantation, we developed a composite implant consisting of MFs embedded within a fibrin hydrogel. Our findings showed that the inclusion of MFs in the hydrogel inhibited fibrin degradation, promoted early recruitment of immune cells, and facilitated axonal regeneration over a 3-month period.
