Universal nanosonosensitizer for ROS-mediated reduction of various cancer cells

Summary: Sonodynamic therapy (SDT) is a promising noninvasive treatment for cancer due to its deep tissue penetration, uniform energy distribution, and external triggers with high specificity and controllability. In this study, four MWO4-PEG nanoparticles (NPs) were synthesized and found to effectively produce reactive oxygen species (ROS) under ultrasound stimulation, leading to decreased cell viability, increased apoptosis rate, and tumor destruction. These NPs showed good sonosensitivity and low toxicity to multiple cancer cell types. Furthermore, NiWO4-PEG, one of the NPs, could be metabolized by the body without long-term toxicity. Overall, these PEGylated MWO4-PEG NPs provide a cost-effective, convenient, and noninvasive approach for universal ultrasound-triggered cancer therapy.

Nanosonosensitizer-Augmented Sonodynamic Therapy Combined with Checkpoint Blockade for Cancer Immunotherapy

Summary: Sonodynamic therapy (SDT) and checkpoint blockade immunotherapy are promising treatments for solid cancers, but their efficacy can be limited. By combining SDT with immunotherapy and nanoparticles, such as TiO2, Chlorin e6, and CpG ODN, a multifunctional nanosonosensitizer was developed to enhance antitumor efficacy of anti-PD-L1 antibody. The nanosonosensitizers not only effectively inhibited tumor growth but also stimulated immune responses, leading to efficient primary tumor growth inhibition and inhibition of distant tumors by inducing a strong tumor-specific immune response. This innovative approach offers a promising strategy for treating malignant tumors.
