It was found that replacing the methoxyl group of colchicine with amino groups increased its selective tubulin binding affinity, G2/M arrest, and antiproliferation activity. Methylaminocolchicine showed stronger antitumor effects compared to paclitaxel and vincristine, without cross-resistance to paclitaxel. It binds to tubulins in unique patterns, inhibits P-gp, and has a stable pharmacokinetic profile.

Summary: A series of pyrimidine-bridged CA-4 derivatives were designed, synthesized, and evaluated for their anti-cancer properties. The compound 9j showed potent anti-proliferative activities against cervical cancer cells and exhibited superior activity against cisplatin-resistant cells. In combination with cisplatin, 9j demonstrated tumor suppression and reduced renal dysfunction. The druggability characterization confirmed that 9j has promising potential as a safe combination agent for cervical cancer therapy.

Summary: Cancer is a leading cause of death globally, and despite advancements in discovering new anti-cancer candidates, it continues to result in numerous deaths. Tubulin protein, which plays a critical role in cell division and intracellular transportation, has become one of the main targets for anti-cancer agents. Inhibiting microtubule formation by targeting tubulin protein can induce cell death. Recent studies have focused on designing and synthesizing novel compounds that target tubulin and have demonstrated anticancer activity against various cancer cell lines. Tubulin inhibitors have shown promising results in inhibiting cancer cell viability, inducing apoptosis, and effectively binding with the colchicine binding site of tubulin.

Summary: In this study, design, synthesis, and evaluation of DHPMs-cinnamic acids hybrids as potential anti-cancer compounds were reported. The results indicated that the hybridization was effective, leading to compounds with much more cytotoxicity than DHPMs and cinnamic acids evaluated separately. Compound 3a was the most potent hybrid and further optimization led to the discovery of three new analogues.
