Multi-organelle-targeting pH-dependent NIR fluorescent probe for lysosomal viscosity

Summary: The normal operation of lysosome, mitochondria, Golgi apparatus, and endoplasmic reticulum is crucial for maintaining cell homeostasis. A new NIR pH-dependent fluorescent probe, DCIC, was synthesized using the TICT mechanism to identify the viscosity of lysosomes. DCIC exhibited high sensitivity to viscosity in viscous media, with fluorescence intensity increasing by 180 times as viscosity increased from 1.0 cp to 438.4 cp. Moreover, DCIC showed a high localization ability for lysosome, mitochondria, Golgi apparatus, and endoplasmic reticulum, and could monitor lysosomal viscosity fluctuations using laser confocal microscopy.

Dual-response near-infrared fluorescent probe for detecting cyanide and mitochondrial viscosity and its application in bioimaging

Summary: This study developed a dual-response coumarin-based fluorescent probe (CCB) for simultaneous detection of mitochondrial viscosity and cyanide (CN-). CCB exhibited a strong log-linear relationship with viscosity, with a correlation coefficient of 0.997. Colorimetric method with a low detection limit of 0.22 μM was employed for CN- detection. Test strips for CN- detection in plants were prepared. The biocompatibility of CCB was further demonstrated, enabling real-time detection of exogenous CN- and intra-cellular mitochondrial viscosity, as well as in vivo bioimaging of viscosity in mice. The probe is crucial for understanding disorders related to mitochondrial viscosity and identifying CN- in daily living.

Mitochondria-targeted fluorescent probe for imaging viscosity in hepatic ischemia-reperfusion injury cell model

A fluorescent probe X-V was designed to detect viscosity changes in a hepatic ischemia-reperfusion injury (HIRI) cell model with high selectivity and sensitivity. A retrospective study conducted in the US examined the effects of the pandemic on parental ratings of concerns related to family conflict, social interaction opportunities, and loss of institutional support. The study found that parents reported increasing levels of concern for themselves and their children over time, with parents' personal concerns rated higher than their child's concerns. Parents of children with co-occurring intellectual disability and younger children reported higher levels of concerns. The study highlights the need for improved service delivery and support for these families.
