Microbial source tracking to elucidate the impact of land-use and physiochemical water quality on fecal contamination in a mixed land-use watershed

Summary: In this study, molecular techniques were used to track the sources of fecal contamination in a mixed land-use watershed in the Appalachian Mountains. Results showed that human-related sources were primarily found in developed areas, indicating that septic or sewer failure is a major source of fecal input. Additionally, it was found that acid mine drainage may contribute to the inactivation or loss of culturability in E. coli. Identifying fecal contamination sources and understanding the impact of in-stream physiochemistry can help develop sustainable watershed management plans to protect water sources.

Comparison of nitrogen losses by runoff from two different cultivating patterns in sloping farmland with yellow soil during maize growth in Southwest China

Summary: The loss of nitrogen in farmland is a significant contributor to agricultural non-point source pollution, affecting the aquatic environment. This study examined the characteristics of runoff and nitrogen losses under different tillage practices over a two-year period. It was found that as rainfall increased, both surface runoff and interflow increased. The surface runoff and nitrogen losses were significantly higher in downslope ridge planting compared to cross ridge planting. The interflow volumes and nitrogen losses in the soil layers of cross ridge planting were also higher than those of downslope ridge planting. Surface runoff was identified as the main pathway for nitrogen losses. Dissolved total nitrogen was the dominant form of nitrogen loss, accounting for the majority of total nitrogen losses.

Cage transplant experiment shows weak transport effect on relative abundance of fish community composition as revealed by eDNA metabarcoding

Summary: The study found that the use of environmental DNA (eDNA) metabarcoding can accurately assess the composition and spatial distribution of freshwater fish communities. In a transplant experiment in the St. Lawrence River, the eDNA of brown trout was detected at five locations up to 5,000 meters from the cage, but only one sampling location downstream from the cage showed a significant impact on community composition. Cluster analysis confirmed the impact of the brown trout eDNA on community composition, separating this location from others. No evidence of transport effect of brown trout eDNA on fish community composition was observed in other sampling locations.
