Differential dynamic microscopy for the characterization of polymer systems

Roberto Cerbino et al.

Summary: DDM is a rapidly emerging approach that transforms a commercial microscope by combining real-space information with the powerful capabilities of conventional light scattering analysis. Analysis of a single microscope movie using DDM gives access to sample dynamics in a wide range of scattering wave-vectors, enabling contemporary polymer science experiments that would be difficult or impossible with standard light scattering techniques. Further applications of DDM to a variety of polymer systems are likely to be just around the corner, making DDM a tool of choice for modern experimental polymer scientists.

Microtubules tune mechanosensitive cell responses

Shailaja Seetharaman et al.

Summary: Mechanotransduction is a process where cells sense mechanical properties of their environment, with integrin-mediated focal adhesions being crucial sites. The crosstalk between microtubules and actin in mechanotransduction affects cell adhesion and migration. Substrate-rigidity-dependent microtubule acetylation controls YAP translocation, focal adhesion distribution, actomyosin contractility, and cell migration.

Extensile to contractile transition in active microtubule-actin composites generates layered asters with programmable lifetimes

John Berezney et al.

Summary: This study investigates a reconstituted composite system composed of an active microtubule network and a passive F-actin filament network. By increasing the concentration of actin filaments, the system's dynamics transition from turbulent-like flows to bulk contractions. At intermediate concentrations, the composite separates into layered asters that coexist with a background turbulent fluid. These structures self-organize and have a defined lifetime.

Encapsulated actomyosin patterns drive cell-like membrane shape changes

Yashar Bashirzadeh et al.

Summary: Cell shape changes during locomotion and cytokinesis are primarily driven by myosin-induced remodeling of cortical actin patterns. The architecture of actin networks, which largely determines the shape of the membrane, is influenced by passive crosslinkers such as alpha-actinin and fascin, as well as the actin nucleator Arp2/3 complex. By reconstituting actomyosin networks in cell-sized lipid bilayer vesicles, the study shows that the size of the vesicle and the concentrations of alpha-actinin and fascin can lead to the assembly of different actomyosin patterns, such as rings and aster-like structures.

Crowding and confinement act in concert to slow DNA diffusion within cell-sized droplets

Mehdi Shafiei Aporvari et al.

Summary: This study uses differential dynamic microscopy to investigate the diffusion of large DNA molecules confined in cell-sized droplets and crowded by dextran polymers. The results show that confined and crowded DNA molecules exhibit universal anomalous subdiffusion, with the degree of confinement and crowding having minimal impact. However, as droplet size decreases, the effective DNA diffusion coefficients decrease significantly, and this effect is enhanced by increased crowding.

Self-repair protects microtubules from destruction by molecular motors

Sarah Triclin et al.

Summary: The study demonstrates that molecular motors generate mechanical energy by removing tubulin dimers and rapidly destroying microtubules, but this damage is compensated for by a self-repair mechanism, allowing microtubules to survive as molecular motors move along their tracks. This reveals the existence of coupling between molecular motor motion and microtubule lattice renewal.

A design framework for actively crosslinked filament networks

Sebastian Furthauer et al.

Summary: Living matter moves, deforms, and organizes itself through polymer filaments and crosslinking molecules in cells. The theory presented predicts the material properties of highly crosslinked filament networks based on the forces exerted by microscopic agents. This work enhances the understanding of cytoskeletal mechanics and paves the way for designing cytoskeletal networks with desired properties in the laboratory.

Multiscale Microtubule Dynamics in Active Nematics

Linnea M. Lemma et al.

Summary: The study quantified the motion of microtubule bundles, revealing that the extension speed of isolated bundles is faster than that in dense active nematics. In the dense active nematic, the sliding speeds of filaments are widely distributed, showing both contractile and extensile relative motion.

Fractal generation in a two-dimensional active-nematic fluid

Kevin A. Mitchell et al.

Summary: Active fluids composed of self-propelled agents can generate complex large-scale coherent flows. Laboratory realization of such fluids with microtubules driven by motor proteins exhibits chaotic advection and pronounced fractal structures. The power spectrum decays as k(-beta) for large wavenumbers in these systems, and a mathematical model based on local compressibility and periodic injection of density variations reproduces this behavior.

Motor-Driven Restructuring of Cytoskeleton Composites Leads to Tunable Time-Varying Elasticity

Janet Y. Sheung et al.

Summary: In this study, light-activated myosin II motors were used to drive composites with varying actin-tubulin molar percentages, showing an increase in elastic plateau modulus and active restructuring of the cytoskeleton components. Nonlinear microrheology measurements demonstrated that motor-driven restructuring enhanced force response and stiffness while suppressing actin bending. The 50-50 composite exhibited the most dramatic mechanical response to motor activity due to a combination of added stiffness from microtubules and sufficient motor substrate.

Active cytoskeletal composites display emergent tunable contractility and restructuring

Gloria Lee et al.

Summary: This study combines microscale experiments with mechanistic modeling to investigate how connectivity, rigidity, and force-generation affect emergent material properties in composite networks of actin, tubulin, and myosin in the cytoskeleton. The research reveals that percolated actomyosin networks are essential for contraction, and a critical fraction of microtubules is necessary to sustain controlled dynamics. The comprehensive phase map presented not only provides important insight into different routes the cytoskeleton can use to alter its dynamics and structure, but also serves as a blueprint for designing cytoskeleton-inspired materials with tunability, resilience, and adaptability for diverse applications.
