Focal thalamus pathology in frontotemporal dementia: Phenotype-associated thalamic profiles

Mary Clare McKenna et al.

Summary: Different clinical subtypes of frontotemporal dementia show distinct thalamic pathology and selective degeneration of specific thalamic nuclei, suggesting thalamic degeneration plays a role in the heterogeneity of FTD manifestations. Utilizing a multimodal approach with complementary MRI techniques can provide a comprehensive understanding of thalamic pathology in FTD.

Thalamic and Cerebellar Regional Involvement across the ALS-FTD Spectrum and the Effect of C9orf72

Martina Bocchetta et al.

Summary: Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) are part of the same disease spectrum. This study used MRI to compare thalamic and cerebellar changes in ALS, FTD, and ALS-FTD patients with and without C9orf72 expansions. The results showed distinct atrophy profiles in the thalamus and cerebellum across the ALS-FTD spectrum, with differential impacts on behavior and cognition. C9orf72 expansions were found to contribute uniquely to the clinical profiles of these patients.

Unravelling the clinical spectrum and the role of repeat length in C9ORF72 repeat expansions

Emma L. van der Ende et al.

Summary: The C9orf72 repeat expansion is associated with a wide range of phenotypes, including different types of dementia, movement disorders, psychiatric symptoms, and slowly progressive FTD. The impact of repeat length on phenotype is unclear, and small expansions may also have clinical significance. Accurate measurement of repeat expansions is hindered by technical difficulties and inconsistent cut-offs between laboratories.

The anterior thalamic nuclei and cognition: A role beyond space?

Andrew J. D. Nelson

Summary: The anterior thalamic nuclei play a crucial role in spatial learning and memory, but recent research has shown that their functions extend beyond spatial processing to include temporal discrimination and processing of other contextual information. Additionally, these nuclei are also important for non-spatial learning, including attentional mechanisms. This indicates the need to reconsider the functions of the anterior thalamic nuclei within a broader framework of connections with sites supporting non-spatial functions such as the anterior cingulate cortex.

Summary: Patients with amyotrophic lateral sclerosis (ALS) with C9orf72 expansion (ALSC9+) have significantly different neuropsychological profiles than those without expansion (ALSC9-), showing more severe impairments in executive functions and verbal memory domains.

Segmental involvement of the corpus callosum in C9orf72-associated ALS: a tract of interest-based DTI study

Hans-Peter Muller et al.

Summary: DTI analysis revealed regional differences in white matter integrity between ALS patients with C9orf72 expansions and sporadic ALS patients, with reductions correlated with clinical parameters. This suggests a specific imaging signature of C9orf72-associated overlap of motor neuron disease and frontotemporal pathology.

Correlation between C9ORF72 mutation and neurodegenerative diseases: A comprehensive review of the literature

Xingfeng Xu et al.

Summary: C9ORF72 encodes a 54-kDa protein with unknown function that is highly expressed in the central nervous system. Its hexanucleotide amplification is associated with neurodegenerative diseases, particularly ALS/FTD spectrum diseases. The causative role of C9ORF72 in AD and PD is still uncertain.
