Affinity binding of COVID-19 drug candidates (chloroquine/ hydroxychloroquine) and serum albumin: Based on photochemistry and molecular docking

Summary: Chloroquine and hydroxychloroquine show effectiveness in the early stage of SARS-CoV-2 treatment, but are no longer recommended due to side effects. The affinity behavior between these drugs and serum albumin was investigated through biochemistry and computer simulation. The results reveal that the drugs quench the intrinsic emission of serum albumin through hydrogen bonding and van der Waals forces, with weak binding constants suggesting differences in efficacy and side effects. Drug modifications can be guided by combining experimental and simulation results to develop CQ derivatives with fewer side effects for COVID-19 treatment.

Bionic lipoprotein loaded with chloroquine-mediated blocking immune escape improves antitumor immunotherapy

Summary: Tumor immunotherapy has great potential for eliminating tumors, but immune escape and the immunosuppressive microenvironment of tumors often limit its efficacy. CD47 on cancer cells can bind to signal regulatory protein alpha (SIRPα) on macrophages and send a "don't eat me" signal, which is an important pathway of immune escape. Additionally, the presence of M2-type macrophages in tumor microenvironment contributes to the immunosuppressive microenvironment. This study presents a drug loading system (BLP-CQ-aCD47) that can block immune escape and improve the immunosuppressive microenvironment by delivering chloroquine (CQ) to M2-type macrophages and blocking CD47-SIRPα interaction. Overall, BLP-CQ-aCD47 provides a new approach for tumor immunotherapy.

Effect of hydroxychloroquine sulfate on the gelation behavior, water mobility and structure of gelatin

Summary: The study found that with increasing HCQ content, the gelation properties of gelatin decreased, as the addition of HCQ led to the formation of hydrogen bonds that interfered with the formation of triple helices in gelatin. FTIR and XRD analysis confirmed that the interaction between HCQ and gelatin is primarily through hydrogen bonding. AFM revealed that higher content of HCQ resulted in more and larger aggregates in gelatin.

Multi-spectroscopic, molecular docking and molecular dynamic simulation evaluation of hydroxychloroquine sulfate interaction with caseins and whey proteins

Summary: This study investigated the interaction between hydroxychloroquine sulfate (HCQ) and caseins (CAS) or whey proteins (WP) using spectroscopic, molecular docking, and molecular dynamic simulation methods. The results showed the formation of HCQ-CAS and HCQ-WP complexes through fluorescence spectrum analysis. Fourier transform infrared spectroscopy (FTIR), circular dichroism spectroscopy (CD), X-ray diffraction (XRD), and scanning electron microscope (SEM) confirmed the formation of these complexes. The study also revealed that HCQ binding altered the secondary structure of CAS and WP. Molecular docking and MD simulation analysis indicated that hydrophobic interactions, hydrogen bonding, and electrostatic attraction forces played a role in the interaction between HCQ and CAS/WP. This study provides insights into using CAS and WP as a drug delivery system for HCQ and enhances our understanding of how HCQ interacts with milk proteins.

Collagen and its derivatives: From structure and properties to their applications in food industry

The limitations and challenges of applying collagen and its derivatives in the food industry are discussed, along with possible research directions to address these challenges.

NMR Investigation of the Interaction of Three Non-Steroidal Anti-Inflammatory Drugs with Human Serum Albumin

Summary: Understanding the interaction between non-steroidal anti-inflammatory drugs and human serum albumin is crucial for the development of new drugs and therapeutic strategies. In this study, high-field solution Nuclear Magnetic Resonance (NMR) spectroscopy was used to compare the strength of interaction of three drugs with albumin. Results showed that diclofenac had a higher affinity index compared to ketorolac and flurbiprofen. Aromatic moieties and methine protons at chiral centers were found to be more involved in the interaction.
