Targeting cellular senescence with senotherapeutics: senolytics and senomorphics

Summary: The concept of geroscience focuses on targeting the ageing process itself as a means to improve human health. Cellular senescence, a state of irreversible cell arrest, has emerged as a potential therapeutic target to extend healthspan. Senescent cells develop a secretory phenotype that promotes chronic inflammation and tissue dysfunction. Therapeutic drugs that induce selective cell death of senescent cells or suppress the expression of senescence markers are being developed.

Summary: This retrospective study evaluated the diagnostic efficiency of hybrid PET/MRI with PSMA in detecting skeletal metastases in newly diagnosed PCa patients. The results showed that the combined PET/MRI evaluation changed the clinical impact in 13.5% of patients and PSMA PET had higher sensitivity while MRI had higher specificity and PPV. Adding WB MRI sequences to PSMA PET imaging appears beneficial for PCa patient management.

Whole-body CDS+ T cell visualization before and during cancer immunotherapy: a phase 1/2 trial

Summary: Immune checkpoint inhibitors (ICIs), by reinvigorating CD8(+) T cell mediated immunity, have revolutionized cancer therapy. However, the systemic distribution and pharmacodynamics of CD8(+) T cells in response to ICIs remains poorly understood. In this study, we used a zirconium-89-labeled, CD8-specific antibody PET tracer to characterize the dynamic distribution of CD8(+) T cells in patients with solid tumors before and after ICI therapy. The results revealed significant heterogeneity in CD8(+) T cell distribution and pharmacodynamics both between and within patients.

Circulating DNA and Liquid Biopsies in the Management of Patients with Cancer

Summary: The study discovered that cancer patients had increased levels of cfDNA in their serum compared to healthy individuals, with the highest levels found in patients with metastatic cancer. Changes in serum DNA levels were found to be correlated with treatment outcomes, with increased levels indicating poor response and decreases indicating a better prognosis. Liquid biopsies based on cfDNA have become an important tool for personalized cancer treatment, allowing for early detection, improved staging, real-time monitoring of therapeutic efficacy, and detection of therapeutic targets and resistance mechanisms.
