Visible-Light-Promoted Cyanoalkylation/Cyclization Cascade Reaction to Assemble Polyheterocycles in Continuous Flow

Summary: This study discusses a visible-light-induced cascade reaction that allows the preparation of cyanoalkyl-containing polyheterocycles. The reaction is initiated by the photoinduced radical cascade addition of N-arylacrylamide derivatives using cyclic oxime esters as radical sources, followed by cyanoalkyl-mediated cyclization. This protocol demonstrates excellent compatibility with different functional groups and provides a variety of desired phenanthridine derivatives in moderate to good yields. Additionally, the use of a microflow technique reduces the reaction time to 10 min compared to traditional batch reactions.

TBADT-Mediated C-C Bond Formation Exploiting Aryl Aldehydes in a Photochemical Flow Reactor

The process was successfully scaled up to produce over 500 kg of the desired intermediate, with the implementation of fully continuous manufacturing.

Preliminary mechanistic experiments suggest a radical-radical cross-coupling mechanism in the reaction.

Furthermore, using photocatalytic continuous flow technique improved the reaction yields, reduced reaction times, and allowed for easy scale-up, highlighting its potential as a synthetic platform.
