The immunogenomic landscape of resected intrahepatic cholangiocarcinoma

Summary: This study integrated immunohistochemistry with next-generation sequencing to analyze intrahepatic CCA samples and found that T-cell and immune checkpoint markers were enriched at tumor margins compared to the center. High PD-1 or LAG-3 and low CD3/CD4/ICOS in the tumor center were associated with poor survival. Loss-of-function mutations in BRCA1-associated protein-1 were associated with elevated expression of the immunosuppressive checkpoint marker B7-H4.

Biomarkers for response to immunotherapy in hepatobiliary malignancies

Summary: The clinical benefit of immunotherapy for hepatobiliary malignancies is limited to a small subgroup of patients, and reliable predictors of immunotherapeutic response are urgently needed. Biomarkers for immunotherapy response in hepatobiliary malignancies lack compelling evidence, with conflicting results on tumor programmed death-ligand 1 (PD-L1) expression and generally low tumor mutational burden (TMB). Emerging biomarkers such as gut microbiota and liquid-biopsy biomarkers show predictive potential.
