Roles of miR-124-3p/Scd1 in urolithin A-induced brown adipocyte differentiation and succinate-dependent regulation of mitochondrial complex II

Qian Li et al.

Summary: In this study, the researchers found that urolithin A (UA) can induce the expression of miR-124-3p, increasing the thermogenic activity of brown adipocytes. miR-124-3p enhances thermogenesis by affecting fatty acid synthesis, specifically targeting the enzyme SCD1, which leads to increased mitochondrial activity and succinate accumulation. This study highlights the important role of the miR-124-3p/SCD1/succinate pathway in regulating thermogenesis of brown adipocytes.

Blueberry juice augments exercise-induced neuroprotection in a Parkinson's disease model through modulation of GDNF levels

Sandra L. Castro et al.

Summary: Exercise combined with consumption of blueberry juice shows greater efficacy in protecting against Parkinson's disease. In animal experiments, the combination of exercise and blueberry juice can reduce behavioral deficits and neurodegeneration to a greater extent compared to either treatment alone. These results suggest that polyphenols may enhance the protective effects of exercise, and differential regulation of neurotrophic factors may be involved.

Combinatorial epigenetic mechanisms of sulforaphane, genistein and sodium butyrate in breast cancer inhibition

Manvi Sharma et al.

Summary: Dietary phytochemicals have the ability to prevent cancer by regulating the epigenome. Some natural botanicals, when combined at optimal concentrations, can synergistically decrease cellular viability and inhibit tumor growth in breast cancer cell lines. These combinations also down-regulate the levels of enzymes involved in epigenetic modifications and induce global changes in chromatin modifications. This study suggests that combined use of these dietary compounds may have implications in breast cancer therapy.

Redox signaling at the crossroads of human health and disease

Jing Zuo et al.

Summary: Redox biology is crucial in life sciences, as redox processes are closely associated with biological activities. Redox homeostasis is essential for human health, where nonradical reactive oxygen species (ROS) function as key second messengers in modulating physiological redox signaling. However, excessive ROS accumulation, termed oxidative stress (OS), leads to biomolecule damage and the development of various diseases. Understanding the roles of redox signaling networks under physiological and pathological conditions is important for the development of redox medicine and treatment of redox-relevant diseases.

Pterostilbene leads to DNMT3B-mediated DNA methylation and silencing of OCT1-targeted oncogenes in breast cancer cells

Megan Beetch et al.

Summary: The study investigated the dynamics between oncogenic TF OCT1 binding and de novo DNA methyltransferase DNMT3B binding in PTS-treated invasive breast cancer cells. It was found that PTS led to decreased OCT1 binding and increased DNMT3B binding in gene regulatory regions associated with oncogenic functions, resulting in gene methylation and silencing. Our findings provide insights into the mechanistic players and gene targets that contribute to the anticancer action of stilbenoid polyphenols.

Summary: Epigenetics refers to DNA chemistry changes that affect gene transcription and translation independently of the DNA coding sequence. These modifications involve various molecular mechanisms, including classic epigenetic changes like DNA methylation and histone modifications, as well as small RNA-mediated processes. Epigenetic changes play a critical role in gene regulation and are reversible and interconnected. They are influenced by genetic predisposition, environmental factors, and lifestyle choices, and are implicated in the development of various diseases. This review focuses on how epigenetic changes are associated with metabolic syndrome and explores strategies such as epigenetic diet, drugs, editing tools, and miRNA-based therapies to modify its outcomes.
