Ruxolitinib Versus Best Available Therapy for Polycythemia Vera Intolerant or Resistant to Hydroxycarbamide in a Randomized Trial

Claire N. Harrison et al.

Summary: The MAJIC-PV study shows that ruxolitinib treatment benefits PV patients resistant or intolerant to hydroxycarbamide (HC-INT/RES) with superior complete response rate, event-free survival, and molecular response; importantly, it also demonstrates for the first time, to our knowledge, that molecular response is linked to event-free survival, progression-free survival, and overall survival.

Clonal Hematopoiesis and Its Impact on Human Health

Herra Ahmad et al.

Summary: Aging is associated with increased mutational burden in all tissues. This can lead to stem cell clonal expansion, which is seen in various tissues but best studied in blood. Clonal hematopoiesis is linked to an increased risk of blood cancers and nonmalignant diseases like cardiovascular disease. Understanding clonal hematopoiesis can provide insights into age-related changes in stem cell composition and function.

Aging of human hematopoietic stem cells is linked to changes in Cdc42 activity

Amanda Amoah et al.

Summary: This study characterizes age-related phenotypes of human hematopoietic stem cells (HSC). It reveals increased frequencies of HSC, hematopoietic progenitor cells, and lineage negative cells in the elderly, as well as a decreased frequency of multi-lymphoid progenitors. The elevated activity of Cdc42 in aged HSC contributes to age-related changes and offers a potential target for attenuating aging in human HSC.

Hematopoietic Stem Cell Heterogeneity Is Linked to the Initiation and Therapeutic Response of Myeloproliferative Neoplasms

Jingyuan Tong et al.

Summary: This study demonstrates that in JAK2V617F(+) ET patients, the Mk-primed HSC subpopulation significantly expands with enhanced potential, and during treatment, mutant HSCs are preferentially targeted in this subpopulation. Homozygous mutant HSCs are forced to re-enter quiescence, while their heterozygous counterparts undergo apoptosis.

Decline in IGF1 in the bone marrow microenvironment initiates hematopoietic stem cell aging

Kira Young et al.

Summary: As individuals age, the function of hematopoietic stem cells (HSCs) declines, leading to limited health span of the blood and immune systems. Research indicates that the hallmarks of aging in HSCs and hematopoiesis start accumulating by middle age, with decreased levels of the longevity-associated molecule IGF1 in the bone marrow microenvironment playing a key role in causing HSC aging. By directly stimulating middle-aged HSCs with IGF1, molecular and functional hallmarks of aging can be rescued, highlighting the complex relationship between IGF1 levels, longevity, and HSC function.
