Synthetic fibrous hydrogels as a platform to decipher cell-matrix mechanical interactions

Summary: Cells sense external forces from the extracellular matrix (ECM) and generate contractile forces to stiffen and remodel the matrix. However, the underlying mechanisms of this bidirectional mechanical exchange are still poorly understood. This study uses a synthetic hydrogel based on polyisocyanide (PIC) polymers to investigate cell-matrix interactions and demonstrates the potential of PIC gels in studying mechanobiology and designing materials.

However, clinical translation of soluble bFGF-based therapies is unsuccessful due to high doses and adverse effects. This study demonstrates the potential of simple biological conjugation procedures to induce additional bioactivity in synthetic cell culture matrices.

Surgical repair aims to create a load-bearing functional pelvic floor by activating fibroblasts to regulate collagen metabolism without fibrotic tissue formation. Fibroblast function plays a crucial role in the pathophysiology of POP and determines the success of treatment.

The model successfully differentiated fibroblasts and myofibroblasts and was used to test the effects of antifibrotic drugs.

Polyisocyanides as a substrate to trigger vaginal fibroblast functioning in an in vitro model for prolapse repair

Summary: Pelvic organ prolapse (POP) is a condition where the bladder, uterus, and/or rectum descend into the vagina. It is associated with changes in vaginal fibroblast functionality and connective tissue composition. Surgical intervention for POP has poor outcomes, possibly due to the lack of true connective tissue restoration. A novel treatment approach involves using a bioactive supplement to enhance tissue repair post-surgery. In this study, the effects of introducing cell binding peptides on a biomimetic gel were investigated using primary vaginal fibroblasts from a POP patient. The results showed that the modified gel promoted cell spread and proliferation, and led to increased production of collagen and elastin by the fibroblasts. These findings suggest that this gel application may improve connective tissue healing in the pelvic floor, which has important implications for regeneration in surgery.

Fabrication of Drug-Eluting Polycaprolactone/poly(lactic-co-glycolic Acid) Prolapse Mats Using Solution-Extrusion 3D Printing and Coaxial Electrospinning Techniques

Summary: Degradable drug-eluting prolapse mats were developed using solution-extrusion 3D printing and coaxial electrospinning techniques. The mats, composed of PCL mesh and PLGA nanofibers with incorporated drugs, exhibited comparable mechanical properties to commercial non-degradable polypropylene knitted meshes for POP repair. In vitro release studies showed sustained release of drugs from the membranes, with no adverse effects observed in animal tests, indicating potential for POP applications.
