Substrate stiffness controls proinflammatory responses in human gingival fibroblasts

Watcharaphol Tiskratok et al.

Summary: Soft gingiva is often compromised in gingival health, but the underlying biological mechanisms remain unknown. In vitro investigation showed that substrate stiffness influences proinflammatory responses in human gingival fibroblasts. Soft substrate induced proinflammatory responses and inhibition of extracellular matrix (ECM) synthesis in hGFs by inactivating cellular mechanotransduction, highlighting the importance of ECM stiffness in gingival health.

Role of Fibroblasts and Myofibroblasts on the Pathogenesis and Treatment of Pelvic Organ Prolapse

Zeliha Guler et al.

Summary: Pelvic organ prolapse (POP) is a connective tissue disorder that occurs due to damage to the supportive structures of the pelvic floor, causing pelvic organs to descend into the vagina. The impaired function of fibroblasts results in disrupted collagen metabolism and affects tissue mechanics. Surgical repair aims to create a load-bearing functional pelvic floor by activating fibroblasts to regulate collagen metabolism without fibrotic tissue formation. Fibroblast function plays a crucial role in the pathophysiology of POP and determines the success of treatment.

Summary: The driving factors for fibrosis and scar formation are fibroblast differentiation and hampered myofibroblast apoptosis. This study developed a high-throughput in vitro fibrosis model using synthetic polyisocyanide hydrogels, which allowed for measuring contraction and subsequent molecular and cellular analysis. The model successfully differentiated fibroblasts and myofibroblasts and was used to test the effects of antifibrotic drugs.

Distinctive structure, composition and biomechanics of collagen fibrils in vaginal wall connective tissues associated with pelvic organ prolapse

Naiwei Chi et al.

Summary: In this study, the multiscale structure, molecular composition, and biomechanics of collagen fibrils in connective tissues of healthy women and POP patients were analyzed. The correlation between these properties and clinical POP quantification scores was established. The study revealed that collagen characteristics change with the progression of POP, suggesting a link between severe prolapse and altered collagen properties. The findings also indicate that the deterioration of the collagen matrix is synchronized with prolapse progression. These results highlight the potential of using non-invasive clinical tests to quantitatively assess collagen abnormalities and guide personalized treatment for POP patients.

Polyisocyanides as a substrate to trigger vaginal fibroblast functioning in an in vitro model for prolapse repair

Aksel N. Gudde et al.

Summary: Pelvic organ prolapse (POP) is a condition where the bladder, uterus, and/or rectum descend into the vagina. It is associated with changes in vaginal fibroblast functionality and connective tissue composition. Surgical intervention for POP has poor outcomes, possibly due to the lack of true connective tissue restoration. A novel treatment approach involves using a bioactive supplement to enhance tissue repair post-surgery. In this study, the effects of introducing cell binding peptides on a biomimetic gel were investigated using primary vaginal fibroblasts from a POP patient. The results showed that the modified gel promoted cell spread and proliferation, and led to increased production of collagen and elastin by the fibroblasts. These findings suggest that this gel application may improve connective tissue healing in the pelvic floor, which has important implications for regeneration in surgery.

3D Cell Culture: Recent Development in Materials with Tunable Stiffness

Desiree Baruffaldi et al.

Summary: Three-dimensional cell culture systems are believed to better simulate physiological conditions compared to traditional two-dimensional systems. Mechanosensing plays a crucial role in controlling various cell functions, and alterations in extracellular matrix structure due to pathologies can impact tissue stiffness. Development of platforms with modulable stiffness may enhance understanding of cell behavior under mechanical stress stimuli.

Fully absorbable poly-4-hydroxybutyrate implants exhibit more favorable cell-matrix interactions than polypropylene

Chantal M. Diedrich et al.

Summary: The study evaluated the use of poly-4-hydroxybutyrate (P4HB) implants as an alternative to synthetic polypropylene (PP) for addressing pelvic organ prolapse. P4HB implants showed better cellular functions, collagen deposition, and mechanical strength compared to PP. The design of the implant also influenced the behavior of vaginal fibroblasts, with rounded pores in P4HB implants showing the greatest fibroblast attachment and proliferation.

Absorbable Electrospun Poly-4-hydroxybutyrate Scaffolds as a Potential Solution for Pelvic Organ Prolapse Surgery

Kim Verhorstert et al.

Summary: This study investigates the use of electrospun P4HB scaffolds for pelvic organ prolapse (POP) repair, aiming to improve surgical outcomes by mimicking natural extracellular matrix structure, releasing estrogen, and promoting collagen metabolism. The results show that electrospun P4HB scaffolds promote better cell proliferation and ECM deposition compared to knitted P4HB implants, suggesting potential for improved surgical outcomes.
